目的:观察心脑宁胶囊对血管性痴呆(VD)大鼠认知功能及海马区β淀粉样蛋白(β-amyloid peptide, Aβ)和Tau蛋白表达情况的影响,探讨心脑宁胶囊的脑保护作用。方法采用双侧颈总动脉永久结扎法制备VD动物模型,设立假手术组、模型组、心脑宁胶囊治疗组(心脑宁组)。以Morris水迷宫试验检测大鼠的逃避潜伏期以评价认知功能,采用免疫组化法测定大鼠海马区Aβ和Tau蛋白的表达。结果术后各时间点模型组与假手术组相比,大鼠的逃避潜伏期延长(P<0.01),Aβ和Tau蛋白表达明显增加(P<0.01);心脑宁组与模型组相比大鼠的逃避潜伏期明显缩短(P<0.05),Aβ和Tau蛋白表达减少(P<0.01)。结论 VD大鼠海马区Aβ和Tau蛋白表达增加,给予心脑宁胶囊治疗后,海马区Aβ和Tau蛋白的表达减少,在一定程度上改善了VD大鼠的认知功能障碍。

목적:관찰심뇌저효낭대혈관성치태(VD)대서인지공능급해마구β정분양단백(β-amyloid peptide, Aβ)화Tau단백표체정황적영향,탐토심뇌저효낭적뇌보호작용。방법채용쌍측경총동맥영구결찰법제비VD동물모형,설립가수술조、모형조、심뇌저효낭치료조(심뇌저조)。이Morris수미궁시험검측대서적도피잠복기이평개인지공능,채용면역조화법측정대서해마구Aβ화Tau단백적표체。결과술후각시간점모형조여가수술조상비,대서적도피잠복기연장(P<0.01),Aβ화Tau단백표체명현증가(P<0.01);심뇌저조여모형조상비대서적도피잠복기명현축단(P<0.05),Aβ화Tau단백표체감소(P<0.01)。결론 VD대서해마구Aβ화Tau단백표체증가,급여심뇌저효낭치료후,해마구Aβ화Tau단백적표체감소,재일정정도상개선료VD대서적인지공능장애。
Objective To observe the effects of Xinnaoning capsule on behavior and the expression of Aβand Tau protein in hippocampus of vascular dementia (VD) rats, and to study the protective effect of Xinnaoning capsule on brain. Methods The rat models of VD were made by adopting the permanent occlusion of bilateral carotid arteries in SD rats. The rats were divided into three groups: sham operated group, VD model group, Xinnaoning capsule group. Morris wa-ter maze was used to test the cognitive function of these rats. The expression of Aβ and Tau protein in hippocampus was determined with immunohistochemical technique. Results Compared with the sham operation rats, cognitive ability of VD model group decreased significantly (P< 0.01), and the expression of Aβ and Tau protein in hippocampus increased significantly (P< 0.01). Cognitive ability of Xinnaoning capsule group improved significantly (P< 0.05), the expression of Aβand Tau protein reduced significantly (P<0.05), compared with the VD model group. Conclusion The expression of Aβ and Tau protein in hippocampus of VD rats is increased. Xinnaoning capsule can inhibit apoptosis of nerve cells and protect nerve cells through Aβ and Tau protein in hippocampus.Xinnaoning capsule can improve the cognitive function in vascular dementia rats.
