目的:探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭能力的影响。方法:分别通过划痕实验、Transwell小室侵袭实验探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭的影响;明胶酶谱实验研究乌司他丁对人口腔表皮样癌细胞系KB分泌活性MMP-9的影响。结果:划痕实验结果显示:与对照组转移细胞数(130.22±14.99)相比,乌司他丁处理后的KB细胞(69.78±11.79)转移能力明显受到抑制( P<0.05);Transwell小室侵袭结果显示,乌司他丁治疗组 KB细胞侵袭的细胞数目(223.44±28.54)明显低于阴性对照组(501.11±30.75),(P<0.05);明胶酶谱结果显示,乌司他丁处理后的KB细胞活性MMP-9的表达明显降低( P<0.05)。结论:乌司他丁可能通过抑制MMP-9的活性从而抑制口腔癌细胞的转移和侵袭。

Objective:To study the effect of Ulinastatin on cell migration and invasion in human oral epidermoid carcinoma cell line (OECC) KB.Methods:Effect of Ulinastatin on migration and invasion in OECC cell line KB was determined by wound healing assay and Transwell chamber invasion assay, respectively;Gelatin zymography experiments was used to detect the effect of Ulinastatin on MMP-9 expression in OECC cell line KB. Results:Wound healing assay showed that KB cell migration was significantly suppressed by Ulinastatin (69.78 ±11.79) compared with control group(130.22±14.99) (P<0.05); Transwell chamber invasion assay showed that invaded cell number in Ulinastatin group (223.44±28.54) was significantly lower compared to the negative control group(501.11±30.75), P<0.05;Gelatin zymography experimental results showed that MMP-9 expression in KB cell was significantly lower after treatment of Ulinastatin (P<0.05).Conclusion: Ulinastatin may inhibits migration and invasion of KB cell via inhibiting the activity of MMP-9.
