目的:比较同一个体来源正常与炎性牙周膜干细胞的生物学性能。方法:分别分离培养同一个体来源正常牙周膜干细胞(hPDLSCs)及炎性牙周膜干细胞(i-hPDLSCs)。流式细胞仪鉴定两种干细胞表面抗原标记;MTT法、克隆形成率检测并比较两者的增殖能力;茜素红染色及油红O染色检测并比较其分化能力;RT-PCR检测成骨相关基因COL-1、Runx-2、BSP-mRNA及成脂相关基因LPL与PPAR γmRNA的表达水平。结果:i-hPDLSCs及hPDLSCs均具有间充质干细胞特性,i-hPDLSCs增殖能力强于hPDLSCs,但分化能力明显弱于hPDLSCs(P<0.05);两者成脂能力比较无统计学差异(P>0.05)。结论:炎症微环境对牙周膜干细胞的生物学性能有一定的影响。

목적:비교동일개체래원정상여염성아주막간세포적생물학성능。방법:분별분리배양동일개체래원정상아주막간세포(hPDLSCs)급염성아주막간세포(i-hPDLSCs)。류식세포의감정량충간세포표면항원표기;MTT법、극륭형성솔검측병비교량자적증식능력;천소홍염색급유홍O염색검측병비교기분화능력;RT-PCR검측성골상관기인COL-1、Runx-2、BSP-mRNA급성지상관기인LPL여PPAR γmRNA적표체수평。결과:i-hPDLSCs급hPDLSCs균구유간충질간세포특성,i-hPDLSCs증식능력강우hPDLSCs,단분화능력명현약우hPDLSCs(P<0.05);량자성지능력비교무통계학차이(P>0.05)。결론:염증미배경대아주막간세포적생물학성능유일정적영향。
AIM:To compare the biological characteristics of inflammatory periodontal ligament stem cells (i-hPDLSCs)and healthy periodontal ligament stem cells (hPDLSCs)from the same donor.METHODS:i-hP-DLSCs and hPDLSCs were isolated from the same donor.Their colony-forming ability,proliferative capacity,cell sur-face antigens and differentiation ability were evaluated and quantified for statistical analysis.RESULTS:Both stem cells have highly proliferative and differentiative potential.i-hPDLSCs had a significantly higher rate of successive cul-ture and proliferation than hPDLSCs(P<0.05).hPDLSCs had a higher rate of cell osteogenic differentiation than i-hPDLSCs (P<0.05).The adipogenic potential of i-hPDLSCs and hPDLSCs showed no statistical difference.CON-CLUSION:Inflammatory environment influences the biological properties of periodontal ligment stem cells.
