目的:建立苜蓿子中总多糖与总多酚含量的测定方法。方法以葡萄糖、没食子酸为对照品,以蒽酮-硫酸、福林试剂-碳酸钠溶液体系为显色剂,采用可见分光光度法在检测波长582、760 nm 处测定10批苜蓿子中总多糖与总多酚的含量。结果葡萄糖溶液浓度在0.02~0.12 mg/mL、没食子酸溶液浓度为1~7μg/mL (r =0.9995)范围内与吸光度呈良好的线性关系,加样回收率为104.30%、101.46%,RSD 为2.40%、2.90%,总多糖的含量为22.34~53.43 mg/g,总多酚的含量为3.03~4.88 mg/g。结论可见分光光度法简单,准确性、重复性良好,为苜蓿子多糖与多酚的进一步研究奠定了基础。

목적:건립목숙자중총다당여총다분함량적측정방법。방법이포도당、몰식자산위대조품,이은동-류산、복림시제-탄산납용액체계위현색제,채용가견분광광도법재검측파장582、760 nm 처측정10비목숙자중총다당여총다분적함량。결과포도당용액농도재0.02~0.12 mg/mL、몰식자산용액농도위1~7μg/mL (r =0.9995)범위내여흡광도정량호적선성관계,가양회수솔위104.30%、101.46%,RSD 위2.40%、2.90%,총다당적함량위22.34~53.43 mg/g,총다분적함량위3.03~4.88 mg/g。결론가견분광광도법간단,준학성、중복성량호,위목숙자다당여다분적진일보연구전정료기출。
Objective To develop a method for content determination of total polysaccharides and polyphe-nols in Medicago sativa L.seeds.Methods Glucose and Gallic acid was used as a reference substance,by anthrone sulfuric acid and Folin reagent - sodium carbonate solution system as chromogenic agent;the visible spectrophotometry was used to determine the contents of total polysaccharides and polyphenols in 10 batches of Medicago sativa L.seeds by UV at 582 nm and 760 nm.Results The concentration of glu-cose in 0.02-0.12 mg/mL and gallic acid in the concentration of 1-7 g/mL (r =0.999 5)showed a good linear relationship between absorbance range.The average recovery was 104.30%,101.46% with RSD=3.40%,2.90%,the contents of polysaccharides and polyphenols were 22.34 - 53.43 mg/g and 3.03 -4.88 mg/g.Conclusion The method is simple,sensitive and accurate,which can offer a basic data for the development research of the Medicago sativa L.
