目的 研究早期糖尿病大鼠视网膜中热休克蛋白27 (Hsp27)的表达.方法 实验研究.对2013年5~8月在福建省眼科研究所将84只SD大鼠用数字法随机分为两组,糖尿病视网膜病变(DR)组与对照组,建立STZ诱导的糖尿病大鼠视网膜病模型,应用免疫组织化学与逆转录PCR从转录和翻译水平观察Hsp27在糖尿病SD大鼠(1、2、4周)视网膜上表达的变化情况.结果 Hsp27蛋白及mRNA在对照组视网膜中不表达或弱表达,在DR组中表达呈进行性增强,差异有统计学意义(P<0.05),Hsp27蛋白主要表达在视网膜神经节细胞层、内外核层及内界膜等.结论 Hsp27参与早期糖尿病视网膜病缺血缺氧与氧化应激的发病机制,对视网膜与视神经细胞起到内源性的保护作用.

Objective To detect the expression of Hsp27 in the retina of diabetic rats,and to investigate the role of Hsp27 in the pathogenesis of diabetic retinopathy (DR).Methods Eighty-four SD rats were randomly divided into two groups:control group and DR group.Diabetes was induced by streptozotocin (STZ) intraperitoneal injection in DR group.The techniques of immunohistochemical and reverse transcription polymerase chain reaction (RT-PCR) were used to examine the expression of Hsp27 protein and mRNA in retina respectively at 1,2,4 weeks after STZ injection.Results The result of immunohistochemical and RT-PCR showed that the expression of Hsp27 protein and mRNA in rats retina were not found in control group,but were found gradually increased in DR group (P <0.05).Conclusions Enhanced expression of endogenous Hsp27 involves in the ischemia hypoxia and oxidative stress,may play a critical role in the pathogenesis of early stage DR.
