采自福建省规模化猪场的疑似病料进行PRRSV分离鉴定,采用RT-PCR方法扩增PRRSV ORF5全基因并进行序列测定分析。从采集的病料中成功分离2株PRRSV,2株分离毒株ORF5与GenBank中选择的参考毒株比较发现,其核苷酸同源性为82.5%~90.6%,氨基酸同源性为80.6%~91.5%。通过序列进行系统进化分析均显示分离毒株属于欧洲型PRRSV,ORF5基因与LV相比变异较大,应加大对欧洲型PRRSV的监控。

The samples were collected from sick pigs suspected of having PRRS from some large-scale pig farms in Fujian province. The virus isolates were propagated in Marc-145 cells and were determined by RT-PCR and the ORF5 genes were sequenced and analyzed. Two strains of PRRSV were isolated and comparison of the ORF5 genome sequences of different PRRSV isolates showed isolations was 82.5%-90.6% nucleotide sequence homology among them, which resulted in 80.6%-91.5% amino acid sequence homology. Phylogenetic analysis through GP5 showed that the isolated strains belonged to European type PRRSV. European type PRRSV existed in Fujian Province, gene ORF5 sequence of virulent strains of isolations variation was significant comparing with LV. We should increase monitoring of European type PRRSV.
