Capsaicin receptor%TRPV1%Nerve injury and regeneration
目的:探讨辣椒素受体在神经压榨性损伤后再生过程中的作用。方法:采用大鼠坐骨神经压榨性损伤在体模型,分别记数损伤前给予或不给予辣椒素受体拮抗剂的情况下,损伤发生1、2周后的健侧、损伤近端、损伤中端及损伤远端横断面的髓神经纤维数目并进行统计学分析。结果:(1)健侧、损伤近端各组别间无统计学差异(P>0.05);(2)坐骨神经损伤中段的神经轴突数量计数发现:单纯损伤1周与损伤2周相比、AMG517处理+损伤后1周与2周相比、单纯损伤2周与AMG517+损伤2周之间相比,其轴突数量的差异均有统计学意义(P<0.01);采用AMG517处理组大鼠坐骨神经损伤部位的轴突数量明显多于单纯损伤组。(3)在坐骨神经损伤远端,其神经轴突的数量随着损伤时间的延长轴突数量有所增加,尤其是损伤前采用AMG517处理组大鼠损伤远端神经轴突的数量明显增多,差异有统计学意义(P<0.01)。结论:阻断TRPV1功能可促进大鼠坐骨神经压榨性损伤后的修复。
目的:探討辣椒素受體在神經壓榨性損傷後再生過程中的作用。方法:採用大鼠坐骨神經壓榨性損傷在體模型,分彆記數損傷前給予或不給予辣椒素受體拮抗劑的情況下,損傷髮生1、2週後的健側、損傷近耑、損傷中耑及損傷遠耑橫斷麵的髓神經纖維數目併進行統計學分析。結果:(1)健側、損傷近耑各組彆間無統計學差異(P>0.05);(2)坐骨神經損傷中段的神經軸突數量計數髮現:單純損傷1週與損傷2週相比、AMG517處理+損傷後1週與2週相比、單純損傷2週與AMG517+損傷2週之間相比,其軸突數量的差異均有統計學意義(P<0.01);採用AMG517處理組大鼠坐骨神經損傷部位的軸突數量明顯多于單純損傷組。(3)在坐骨神經損傷遠耑,其神經軸突的數量隨著損傷時間的延長軸突數量有所增加,尤其是損傷前採用AMG517處理組大鼠損傷遠耑神經軸突的數量明顯增多,差異有統計學意義(P<0.01)。結論:阻斷TRPV1功能可促進大鼠坐骨神經壓榨性損傷後的脩複。
목적:탐토랄초소수체재신경압자성손상후재생과정중적작용。방법:채용대서좌골신경압자성손상재체모형,분별기수손상전급여혹불급여랄초소수체길항제적정황하,손상발생1、2주후적건측、손상근단、손상중단급손상원단횡단면적수신경섬유수목병진행통계학분석。결과:(1)건측、손상근단각조별간무통계학차이(P>0.05);(2)좌골신경손상중단적신경축돌수량계수발현:단순손상1주여손상2주상비、AMG517처리+손상후1주여2주상비、단순손상2주여AMG517+손상2주지간상비,기축돌수량적차이균유통계학의의(P<0.01);채용AMG517처리조대서좌골신경손상부위적축돌수량명현다우단순손상조。(3)재좌골신경손상원단,기신경축돌적수량수착손상시간적연장축돌수량유소증가,우기시손상전채용AMG517처리조대서손상원단신경축돌적수량명현증다,차이유통계학의의(P<0.01)。결론:조단TRPV1공능가촉진대서좌골신경압자성손상후적수복。
Objective:To observe the effect of the transient receptor potential vanilloid 1 (TRPV1) on neuronal regeneration after crush injury to the rat’s sciatic nerve.Method:The right sciatic nerve of the adult male Sprague-Dawley rats were crushed.Rats in the experimental group received a subcutaneous injection of AMG-517 ,a highly potent and selective TRPV1 antagonist, into the right hind paw 1 h before the crush.At the crush zone, proximal zone and distal zone, the numbers of myelinated fibers on the transverse section were counted after a surviving time of 7 or 14 days. Result:(1)At the intact side and the proximal zone,the numbers of myelinated fibers did not show any difference. (2) the sciatic nerve injury in the middle of neural axon number counting found that damage to 1 week compared with in 2 weeks, AMG517 treatment+1 week after injury compared with 2 weeks, pure injury and compared between AMG517 + damage, its axon number difference had statistical significance (P<0.01).Use AMG517 treatment group rats sciatic nerve injury of axon number significantly more than the simple injury group.(3) In the distal sciatic nerve injury, the nerve axon number of damage extended axon number increased with time, especially the damage before used AMG517 treatment group rats damage distal nerve axon number increased obviously, the difference was statistically significant (P<0.01). Conclusion:Blockage of the TRPV1 can promote neuronal regeneration after sciatic crush injury.
