目的:显示大鼠左侧冠状动脉的走行情况,探讨冠脉显色指导下冠脉结扎法制作大鼠心肌梗死模型在模型死亡率、成功率及稳定性方面的优势。方法:8只SD大鼠处死后取出心脏,行主动脉插管后肝素盐水灌洗冠状动脉,1%、2%、3%、4%不同浓度37℃琼脂糖凝胶行主动脉根部灌注,确定最佳琼脂糖凝胶浓度;101只雄性SD大鼠行琼脂糖凝胶主动脉灌注,显示出大鼠冠状动脉的分布走行情况;分析冠脉走行情况,制定结扎策略,比较高位结扎法、低位结扎法在大鼠存活率、结扎成功率、梗死灶稳定性方面的优劣。结果:2%37℃琼脂糖凝胶主动脉灌注能清楚的显示冠状动脉在心脏表面的走行;大鼠冠脉的走行可见分为4种类型;60只雄性SD大鼠分别按高位结扎、低位结扎的方法能制作稳定的大鼠心梗模型。结论:冠脉显色指导下冠脉结扎法能制作出稳定的大面积和中等面积大鼠心肌梗死模型。
目的:顯示大鼠左側冠狀動脈的走行情況,探討冠脈顯色指導下冠脈結扎法製作大鼠心肌梗死模型在模型死亡率、成功率及穩定性方麵的優勢。方法:8隻SD大鼠處死後取齣心髒,行主動脈插管後肝素鹽水灌洗冠狀動脈,1%、2%、3%、4%不同濃度37℃瓊脂糖凝膠行主動脈根部灌註,確定最佳瓊脂糖凝膠濃度;101隻雄性SD大鼠行瓊脂糖凝膠主動脈灌註,顯示齣大鼠冠狀動脈的分佈走行情況;分析冠脈走行情況,製定結扎策略,比較高位結扎法、低位結扎法在大鼠存活率、結扎成功率、梗死竈穩定性方麵的優劣。結果:2%37℃瓊脂糖凝膠主動脈灌註能清楚的顯示冠狀動脈在心髒錶麵的走行;大鼠冠脈的走行可見分為4種類型;60隻雄性SD大鼠分彆按高位結扎、低位結扎的方法能製作穩定的大鼠心梗模型。結論:冠脈顯色指導下冠脈結扎法能製作齣穩定的大麵積和中等麵積大鼠心肌梗死模型。
목적:현시대서좌측관상동맥적주행정황,탐토관맥현색지도하관맥결찰법제작대서심기경사모형재모형사망솔、성공솔급은정성방면적우세。방법:8지SD대서처사후취출심장,행주동맥삽관후간소염수관세관상동맥,1%、2%、3%、4%불동농도37℃경지당응효행주동맥근부관주,학정최가경지당응효농도;101지웅성SD대서행경지당응효주동맥관주,현시출대서관상동맥적분포주행정황;분석관맥주행정황,제정결찰책략,비교고위결찰법、저위결찰법재대서존활솔、결찰성공솔、경사조은정성방면적우렬。결과:2%37℃경지당응효주동맥관주능청초적현시관상동맥재심장표면적주행;대서관맥적주행가견분위4충류형;60지웅성SD대서분별안고위결찰、저위결찰적방법능제작은정적대서심경모형。결론:관맥현색지도하관맥결찰법능제작출은정적대면적화중등면적대서심기경사모형。
Objective:To show the anatomy of left coronary artery in rats and discuss the advantage of the death rate, success rate and stability of myocardial infarction model through ligating of the coronary artery under staining guidance.Method:8 heart harvested from sacrificed rat and irrigated coronary artery with saline injection after aortic intubation.The best concentration of agarose gel were identified, 1%, 2%, 3% and 4% agarose gel were used to perfuse aortic root at 37 ℃.101 male rats were perfused to show the anatomy of left coronary artery, then made ligation strategies though analyzed coronary artery distribution, and compared the advantages and disadvantages in death rate, success rate and advantage of stability between high and low ligation.Result:It showed the coronary artery clearly though aortic perfusion by 2% agarose gel at 37 ℃, and the coronary anatomy can mainly be classified into 4 groups. We had made stable myocardial infarction model both in high and low ligation.Conclusion:The method of ligation of the coronary artery under staining guidance can produce stable rat myocardial infarction model both in large and middle infarct size.
