目的:研究小枝玫瑰对α-葡萄糖苷酶抑制作用和对正常小鼠血糖的影响。方法手术摘取大鼠小肠段,获得α-葡萄糖苷酶,以4-硝基苯基-α-D-吡喃葡萄糖苷(pNPG)为底物,测定玫瑰花水提液(Ms)、醇提液(Mc)对α-葡萄糖苷酶活性的抑制作用,并选取α-葡萄糖苷酶活性抑制活性较强的 Mc,动物实验验证其降糖活性。结果玫瑰花不同提取液均有一定的α-葡萄糖苷酶抑制活性,其中 Ms 的 IC50为23.14μg/mL,Mc 的 IC50为11.35μg/mL;Mc多次给药能够明显降低正常小鼠血糖,且呈现一定的量效关系,其中 Mc 中、高剂量降血糖作用优于阿卡波糖,表现为较好的抗糖尿病作用。结论玫瑰花醇提液抑制α-葡萄糖苷酶活性作用强于水提液,抑制α-葡萄糖苷酶活性可能是玫瑰花发挥降糖作用的主要作用机制。

목적:연구소지매괴대α-포도당감매억제작용화대정상소서혈당적영향。방법수술적취대서소장단,획득α-포도당감매,이4-초기분기-α-D-필남포도당감(pNPG)위저물,측정매괴화수제액(Ms)、순제액(Mc)대α-포도당감매활성적억제작용,병선취α-포도당감매활성억제활성교강적 Mc,동물실험험증기강당활성。결과매괴화불동제취액균유일정적α-포도당감매억제활성,기중 Ms 적 IC50위23.14μg/mL,Mc 적 IC50위11.35μg/mL;Mc다차급약능구명현강저정상소서혈당,차정현일정적량효관계,기중 Mc 중、고제량강혈당작용우우아잡파당,표현위교호적항당뇨병작용。결론매괴화순제액억제α-포도당감매활성작용강우수제액,억제α-포도당감매활성가능시매괴화발휘강당작용적주요작용궤제。
Objective To establish the evaluation system of the activity ofα-glucosidase inhibition in vitro and to ex-plore the influence of the extracts on blood glucose of normal mice.Methods α-glucosidase was prepared from rat small intestine via operation.4-nitrobenzophenone-α-D-glucopyranoside (pNPG),as a substrate,was used to measure the inhibition ofα-glucosidase activity by Rosa rugosa water extract (Ms)and Rosa rugosa ethanol extract (Mc)respectively.The MG2 with strong inhibition was selected to validate the activity of lowering blood glucose with animal experiments.Results The different kinds of extracts of Rosa rugosa both showed certainα-glucosidase inhibitory activity,among which the IC50 values of Ms and MG2 was 23.14μg/mL and 11.35μg/mL.Mc could re-duce blood glucose of normal mice;Mc multiple dosing can significantly decrease the blood glucose level in normal mice,and showed dose-effect relationship,and high dosage of Mc had much better anti-diabetic effect than that of acarbose.Conclusion The Rosa rugosa ethanol extract had higherα-glucosidase inhibitory activity than that of the water extract,and it’s likely that the inhibitingα-glucosidase activity was the main mechanism that Rosa rugosa was capable of decreasing blood glucose.
