目的：体外探讨脐血间充质干细胞（UCB－MSCs）对系统性红斑狼疮（SLE）患者外周血单个核细胞（PBMNCs）细胞因子分泌的影响。方法①UCB－MSCs的分离、扩增及鉴定；②健康志愿者及SLE患者PBMNCs的分离。③UCB－MSCs与植物凝集素（ PHA）刺激的PBMNCs共培养， ELISA法检测共培养前后各组细胞培养上清液中IL－6、IL－10、IL－2、IL－12、IFN－γ的含量。结果①UCB－MSCs呈梭形贴壁生长，能在体外连续传代，均表达CD13、CD44、CD166，不表达 CD34、CD45；②UCB－MSCs能显著下调 SLE患者 PBMNCs中 IL－10、IL－6的分泌而上调 IL－2、IL－12、IFN－γ的分泌。结论 UCB－MSCs在体外能改变SLE患者的Th1／Th2极化状态。
목적：체외탐토제혈간충질간세포（UCB－MSCs）대계통성홍반랑창（SLE）환자외주혈단개핵세포（PBMNCs）세포인자분비적영향。방법①UCB－MSCs적분리、확증급감정；②건강지원자급SLE환자PBMNCs적분리。③UCB－MSCs여식물응집소（ PHA）자격적PBMNCs공배양， ELISA법검측공배양전후각조세포배양상청액중IL－6、IL－10、IL－2、IL－12、IFN－γ적함량。결과①UCB－MSCs정사형첩벽생장，능재체외련속전대，균표체CD13、CD44、CD166，불표체 CD34、CD45；②UCB－MSCs능현저하조 SLE환자 PBMNCs중 IL－10、IL－6적분비이상조 IL－2、IL－12、IFN－γ적분비。결론 UCB－MSCs재체외능개변SLE환자적Th1／Th2겁화상태。
OBJECTIVE To explore the effect of umbilical cord blood mesenchymal stem cells on the secretion of ctokinesbyperipheralbloodmononuclearcellsfromsystemiclupuserythematosuspatients.METHODS ①The isolation,expansion and cryopreservation of UC-MSCs;②The isolation,purification and cultivation of PBMNCs from SLE patients and normal person.③UCB-MSCs and PBMNCs stimulated by PHA of SLE patients were coculture, ELISA method was used to detect the content of IL-6,IL-10,IL-2,IL-12 and IFN-γin the cell culture supernatants of eachgroupbeforeandafterco-culture.RESULTS ①UCB-MSCsadheredthebottomoftheflaskandwerefusiform shape,could continuously passage in vitro,express CD13,CD44andCD166 but not CD34 and CD45;②UCB-MSCs could down-regulate the secretion of IL-10 and IL-6 of PBMNCs of SLE patients,and up-regulate the secretion of IL-2,IL-12 and IFN-γ.CONCLUSION UCB-MSCs can change the Th1/Th2 polarization in patients with SLE in vitro.
