[目的]观察电针(EA)对完全弗氏佐剂(CFA)诱导的慢性炎性痛大鼠患侧背根神经节(DRG)N-甲基-D-天门冬氨酸受体亚基1(NR1)受体及NR1丝氨酸890位点磷酸化(pNR1 serine-890)的影响。[方法]健康雄性SD大鼠30只,随机分为正常对照组(N组),模型对照组(CFA组),电针组(EA组),每组10只。右后足底注射CFA 0.1ml以建立慢性炎性痛模型,分别测量大鼠造模前,造模后第1d、3d、5d、7d、9d患侧痛阈变化。免疫荧光法检测大鼠患侧DRG中NR1阳性细胞表达率,免疫印迹法检测大鼠患侧DRG中pNR1 serine-890的表达量。[结果]CFA造模后1d,CFA组与EA组足缩腿阈(PWTs)均低于N组(P<0.01),CFA组与EA组差异无统计学意义;治疗3d起,EA组PWTs呈现升高趋势,于第9d明显高于同时点CFA组(P<0.01)。CFA造模后第9d,与N组比较,CFA组大鼠患侧NR1和pNR1表达均有显著上升(P<0.01,P<0.01);与CFA组比较,EA组大鼠患侧NR1和pNR1表达均下降(P<0.01,P<0.01)。[结论]电针能有效拮抗CFA诱导的大鼠慢性炎性痛,此作用可能与电针下调大鼠患测DRG中的NR1表达量以及减少NR1丝氨酸890位点磷酸化相关。

[목적]관찰전침(EA)대완전불씨좌제(CFA)유도적만성염성통대서환측배근신경절(DRG)N-갑기-D-천문동안산수체아기1(NR1)수체급NR1사안산890위점린산화(pNR1 serine-890)적영향。[방법]건강웅성SD대서30지,수궤분위정상대조조(N조),모형대조조(CFA조),전침조(EA조),매조10지。우후족저주사CFA 0.1ml이건립만성염성통모형,분별측량대서조모전,조모후제1d、3d、5d、7d、9d환측통역변화。면역형광법검측대서환측DRG중NR1양성세포표체솔,면역인적법검측대서환측DRG중pNR1 serine-890적표체량。[결과]CFA조모후1d,CFA조여EA조족축퇴역(PWTs)균저우N조(P<0.01),CFA조여EA조차이무통계학의의;치료3d기,EA조PWTs정현승고추세,우제9d명현고우동시점CFA조(P<0.01)。CFA조모후제9d,여N조비교,CFA조대서환측NR1화pNR1표체균유현저상승(P<0.01,P<0.01);여CFA조비교,EA조대서환측NR1화pNR1표체균하강(P<0.01,P<0.01)。[결론]전침능유효길항CFA유도적대서만성염성통,차작용가능여전침하조대서환측DRG중적NR1표체량이급감소NR1사안산890위점린산화상관。
Objective]To observe the effect of electroacupuncture(EA) on N-methyl-d-aspartate receptors 1(NR1) and phosphorylationin of NR1 at serine-890 site in ispilateral dorsal root ganglion(DRG) of rats with chronic inflammatory pain induced by CFA. [Methods]30 adult male Sprague Dawley(SD) rats were randomly divided into 3 groups:normal(N) group, CFA group, EA group, each group 10 rats. The model of chronic inflammatory pain was established by subcutaneously injecting CFA(0.1ml per rat) into the right hind paw. The paw withdrawal thresholds(PWTs) were measured before CFA injection, as well as at 1d, 3rd, 5th, 7th,9th day after CFA injection. The expression of NR1 in ispilateral DRG was detected by immunofluorescence assay and the expression of pNR1 serine-890 in ispilateral DRG was detected by western blot. [Result]At 1d post-injection, the PWTs of CFA group and EA group were significantly lower than N group(P<0.01), the differences between CFA group and EA group didn't have statistical significance. Compared with CFA group, PWTs of EA group showed increasing tendency from 3d post-injection and were obviously higher than CFA group at same time point( P<0.01). Compared with N group, the expression of NR1 and pNR1 serine-890 in ispilateral DRG of CFA rats both increased with obvious significance( P<0.01, P<0.01).Compared with CFA group, expression of NR1 and pNR1 serine-890 in ispilateral DRG of EA rats both reduced with obvious significance( P<0.01, P<0.01). [Conclusion]EA has an obvious analgesic effect on chronic inflammatory pain induced by CFA, the effect is possibly associated with down-regulation of NR1 and phosphorylation of NR1 at serine-890 site in ispilateral DRG.
