目的:探讨氧化苦参碱对HepG2细胞胰岛素抵抗( IR-HepG2)模型的影响。方法采用以棕榈酸为诱导剂建立IR-HepG2细胞模型,给予不同剂量的氧化苦参碱溶液(12.5~100μg? mL -1),分别测定葡萄糖消耗量、己糖激酶活力、丙酮酸激酶活力和肝糖原含量。结果与正常组相比,模型组细胞的葡萄糖消耗量、己糖激酶活力、丙酮酸激酶活力和肝糖原含量均显著降低,表明胰岛素抵抗模型建立成功;与模型组相比,25~100μg? mL -1的氧化苦参碱可显著增加IR-HepG2细胞的葡萄糖消耗量,50~100μg? mL -1的氧化苦参碱可显著提高 IR-HepG2细胞的己糖激酶活力、丙酮酸活力和肝糖原合成量。结论氧化苦参碱可通过增加葡萄糖消耗量、己糖激酶活力、丙酮酸活力和肝糖原合成量改善 IR-HepG2细胞的胰岛素抵抗。

목적:탐토양화고삼감대HepG2세포이도소저항( IR-HepG2)모형적영향。방법채용이종려산위유도제건립IR-HepG2세포모형,급여불동제량적양화고삼감용액(12.5~100μg? mL -1),분별측정포도당소모량、기당격매활력、병동산격매활력화간당원함량。결과여정상조상비,모형조세포적포도당소모량、기당격매활력、병동산격매활력화간당원함량균현저강저,표명이도소저항모형건립성공;여모형조상비,25~100μg? mL -1적양화고삼감가현저증가IR-HepG2세포적포도당소모량,50~100μg? mL -1적양화고삼감가현저제고 IR-HepG2세포적기당격매활력、병동산활력화간당원합성량。결론양화고삼감가통과증가포도당소모량、기당격매활력、병동산활력화간당원합성량개선 IR-HepG2세포적이도소저항。
ABSTRACT:OBJECTIVE To investigate the effect of oxymatrine on insulin-resistance ( IR-HepG2 ) HepG2 cells.METHODS With palmitic acid as inducer of establishment of IR-HepG2 cell model and given different do-ses,Oxymatrine solution of (12.5~100μg? mL-1 ) ,glucose consumption,the activities of hexokinase,pyruvate ki-nase activity and liver glycogen content were determined.RESULTS Compared with the normal group, model group,the glucose consumption of cells,the activities of hexokinase,pyruvate kinase activity and liver glycogen con-tent were significantly reduced,showed that insulin-resistance model was established successfully;compared with the model group,25~100μg? mL-1 of oxymatrine significantly increased the glucose consumption of IR-HepG2 cells,50~100μg? mL-1 of oxymatrine alkali can significantly increase the activities of hexokinase,IR-HepG2 cells activity and liver glycogen synthesis of pyruvic acid.CONCLUTION Oxymatrine can be improved insulin-resistance by in-creasing the glucose consumption of IR-HepG2 cells,the activities of hexokinase,pyruvate kinase activity and liver glycogen synthesis.
