目的::观察NO 在白藜芦醇减轻大鼠肝缺血再灌注损伤中的作用并探讨其机制。方法:30只健康雄性SD大鼠随机分为3组:假手术组(SC 组)、缺血再灌注组(I/R 组)和白藜芦醇后处理组(Res组)。测定各组大鼠再灌注后2 h 血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(AKP)、透明质酸酶(HA)及一氧化氮(NO)含量,及再灌注后2 h 肝组织 NO、总一氧化氮合酶(T-NOS)及诱导型NOS (iNOS)的水平,光镜观察各组肝细胞的显微结构变化。结果:与 SC 组相比,I/R 组血清 ALT、AST、AKP和 HA活性明显增高而NO 水平降低,病理学检查可见肝细胞肿胀,肝窦变窄,嗜中性粒细胞浸润和片状坏死。Res组再灌注后血清 ALT、AST、AKP 和 HA 值明显下降,肝脏病理损伤改善,同时NO 和T-NOS水平升高而iNOS水平降低。结论:Res 可以通过提高 I/R 大鼠体内 NO 和 eNOS,降低iNOS水平,改善肝脏微循环障碍对肝脏起保护作用。

목적::관찰NO 재백려호순감경대서간결혈재관주손상중적작용병탐토기궤제。방법:30지건강웅성SD대서수궤분위3조:가수술조(SC 조)、결혈재관주조(I/R 조)화백려호순후처리조(Res조)。측정각조대서재관주후2 h 혈청곡병전안매(ALT)、곡초전안매(AST)、감성린산매(AKP)、투명질산매(HA)급일양화담(NO)함량,급재관주후2 h 간조직 NO、총일양화담합매(T-NOS)급유도형NOS (iNOS)적수평,광경관찰각조간세포적현미결구변화。결과:여 SC 조상비,I/R 조혈청 ALT、AST、AKP화 HA활성명현증고이NO 수평강저,병이학검사가견간세포종창,간두변착,기중성립세포침윤화편상배사。Res조재관주후혈청 ALT、AST、AKP 화 HA 치명현하강,간장병리손상개선,동시NO 화T-NOS수평승고이iNOS수평강저。결론:Res 가이통과제고 I/R 대서체내 NO 화 eNOS,강저iNOS수평,개선간장미순배장애대간장기보호작용。
Objective:To investigate the role and mechanism of nitric oxide(NO)in protective effects of post-conditioning with resveratrol against hepatic ischemia/reperfusion inj ury in rats.Methods:30 healthy male Sprague-Dawley (SD)rats were randomly divided into 3 groups:sham control (SC)group,ischemic/reperfusion (I/R)group,and post-disposal treatment of resveratrol group(Res).The activity of alanine aminotransferase (ALT),aspartate aminotransferase (AST),alkaline phosphatase (AKP),the content hyaluronic acid(HA) and NO in serum were detected by 2 h reperfusion;The content of nitric oxide (NO),and the activity of total nitric oxide synthase (T-NOS), inducible nitric oxide synthase(iNOS)in hepatic tissues was measured;the change of microstructure of hepatic cell was detected by HE staining.Results:As compared with Sham Group,the activity of ALT,AST,AKP,iNOS and the content of HA were significantly increased in I/R Group.but the content of NO was decreased;Pathological examination showed swelling of liver cells,hepatic sinus narrow,neutrophil infiltration and patchy necrosis.The activity of ALT,AST,AKP,iNOS and the content of HA were significantly decreased in Res Group after reperfusion;the amount of NO,T-NOS were higher in Res Group,however,the amount of was attenuated;The hepatic structure was improved at the same time.Conclusion:Our results show that resveratrol exerts a potent protective effect on early-stage hepatic ischemia reperfusion inj ury in rats by increasing the content of NO derived from eNOS and reducing hepatic microcirculation disturbance.
