应用超高效液相色谱-串联质谱技术,建立了3种中兽药制剂中非法添加阿莫西林和氨苄西林的检测方法。样品经50%乙腈水溶液提取、稀释后进行分析。实验采用0.2%甲酸乙腈溶液与0.2%甲酸水溶液作为流动相进行梯度洗脱,经Atlantis T3柱(2.1 mm×150 mm,5μm)分离,通过多反应监测模式进行测定。结果表明,阿莫西林和氨苄西林在1~50 ng/mL的范围内线性关系良好,最低检测限分别为0.5 mg/g和0.2 mg/g。阿莫西林回收率在83.4%~95.8%之间,RSD为2.0%~4.6%;氨苄西林回收率在85.1%~97.2%,RSD为1.7%~4.0%。本方法准确、可靠,可用于中兽药制剂中非法添加阿莫西林和氨苄西林的检测。

응용초고효액상색보-천련질보기술,건립료3충중수약제제중비법첨가아막서림화안변서림적검측방법。양품경50%을정수용액제취、희석후진행분석。실험채용0.2%갑산을정용액여0.2%갑산수용액작위류동상진행제도세탈,경Atlantis T3주(2.1 mm×150 mm,5μm)분리,통과다반응감측모식진행측정。결과표명,아막서림화안변서림재1~50 ng/mL적범위내선성관계량호,최저검측한분별위0.5 mg/g화0.2 mg/g。아막서림회수솔재83.4%~95.8%지간,RSD위2.0%~4.6%;안변서림회수솔재85.1%~97.2%,RSD위1.7%~4.0%。본방법준학、가고,가용우중수약제제중비법첨가아막서림화안변서림적검측。
A method based on ultra-performance liquid chromatography tandem mass spectrometry ( UPLC-MS/MS) was developed for the determination of amoxicillin and ampicillin illegally added in 3 kinds of traditional Chinese medicine for animals. Samples were extracted by 50% acetonitrile solution and diluted. The analyte was separated by Atlantis T3 column with a linear gradient elution program of 0.2% formic acid acetonitrile and 0.2%formic acid aqueous solution. Multiple reaction monitoring was used for the selective detection. The results demonstrated that the linearity range of amoxicillin and ampicillin were 1~50 ng/mL, and the limit of detection was 0.5 mg/g and 0. 2 mg/g, respectively. The recovery was 83. 4%~95. 8% with RSD of 2. 0%~4. 6% for amoxicillin, and the recovery was 85.1%~97.2% with RSD of 1.7%~4.0% for ampicillin. It indicated that the method was accurate and reliable for determination of amoxicillin and ampicillin illegally added in 3 kinds of traditional Chinese medicine for animals.
