以体外培养的hGCCs MGC-803细胞系为研究对象,采用不同浓度石蒜碱(5、2.5、1.25μmol/L)进行处理,倒置显微镜下观察其生长状态,同时采用血球计数板统计其体外增殖率,采用流式细胞技术检测处于不同细胞周期的细胞比例.结果表明,石蒜碱处理对hGCCs的体外增殖具有一定的抑制作用,其中5μmol/L石蒜碱对hGCCs的抑制作用最为明显.显微观察和计数分析结果表明,5μmol/L石蒜碱处理的hGCCs活力最低,细胞凋亡现象最明显.细胞周期检测的结果显示石蒜碱处理会使hGCCs增殖趋缓,细胞周期延长.

The hGCCs were treated with different concentration of lycorisradiata alkali (5、2.5、1.25 μmol/L re-spectively), growth status were observed by the inverted microscope, and proliferation rate were count by hemocytome-ter, the proportion of hGCCs in different cell cycles were detected by flow cytometry.The results showed that the prolif-eration of hGCCs was suppressed by treated with lycorisradiata alkali, the most obvious suppressed of hGCCs was 5μmol/L treatment group.Through microscopic observation and counting analysis results, it has been found that the vi-tality of hGCCs was the lowest and the cells apoptosis was the most obvious by treated of 5μmol/L lycorisradiata alka-li.The result of cell cycle detection revealed that by treated with lycorisradiata alkali, the reproduction of hGCCs be-came slow and cell cycle became extended.
