目的:探讨支气管哮喘患者诱导痰炎性细胞中 Tim-3 mRNA 的表达及作用机制,并分析其与上清液中 IL-4、IFN-γ和肺功能指标的相关性。方法将2012年5月-2013年8月我院收治的支气管哮喘患者分为哮喘缓解组和哮喘发作组,并于同期随机选取120例健康体检者为对照组,通过逆转录-聚合酶链反应技术(RT-PCR)检测三组的诱导痰炎性细胞中 Tim-3 mRNA 的水平,同时采用酶联免疫吸附法(ELISA)检测上清液中 IL-4、IFN-γ,分析其与 Tim-3 mRNA 的相关性。结果哮喘发作组、哮喘缓解组 FEV1、FVC、FEV1/FVC 低于对照组,哮喘发作组低于哮喘缓解组,差异有统计学意义(P <0.05),哮喘发作组、哮喘缓解组的 Tim-3/β-actin 水平高于健康对照组,且哮喘发作组高于哮喘缓解组,差异有统计学意义(P <0.05);哮喘发作组、哮喘缓解组 IL-4高于健康对照组,哮喘发作组高于哮喘缓解组,哮喘发作组、哮喘缓解组 IFN-γ低于健康对照组,哮喘发作组低于哮喘缓解组,差异有统计学意义(P <0.05);哮喘缓解组 Tim-3 mRNA 与 IL-4有正相关关系(r =0.71,P <0.05),与 IFN-γ和 FEV1/FVC 有负相关关系(r =-0.86、-0.81,P <0.05),哮喘发作组 Tim-3 mRNA 与 IL-4有正相关关系(r=0.65,P <0.05),与 IFN-γ和 FEV1/FVC 有负相关关系(r=-0.77、-0.78,P <0.05)。结论通过检测 Tim-3 mRNA 水平可以有效评估支气管哮喘的发生与发展,临床有重要参考价值。

Objective To investigate the expression and mechanism of inflammatory cells in induced sputum of pa-tients with bronchial asthma and the correlation between IL-4 and IFN-γ in supernatant.Methods The patients with bronchial asthma in our hospital from 2012 May to 2013 August were divide into asthma remission group and asthma group,and 120 of health volunteers as control group were randomly selected in the same period.The Tim-3 mRNA of inflammatory cells in induced sputum was detected by RT-PCR,and the IL-4、IFN-γ were detected by ELISA,and the correlation was analyzed.Results The FEV1、FVC、FEV1/FVC of asthmatic group and asthma remission group were lower than those of control group when its were lower than those in asthma remission group,the difference was statisti-cally significant(P <0.05),Tim-3/β-actin of asthmatic group and asthma remission group were higher than that of con-trol group when it was higher than that in asthma remission group,the difference was statistically significant(P <0.05);The IL-4 of asthmatic group and asthma remission group were higher than those of control group when its were higher than those in asthma remission group,The IFN-γof asthmatic group and asthma remission group was lower than that of control group when it was lower than that in asthma remission group,the difference was statistically significant (P <0.05);There was a positive correlation between Tim-3 mRNA and IL-4(r=0.71,P <0.05)in asthma remission group when a negative correlative with IFN-γand FEV1/FVC(r=-0.86、-0.81,P <0.05),and there was a positive correlation between Tim-3 mRNA and IL-4(r =0.65,P <0.05)in asthmatic group when a negative correlative with IFN-γand FEV1/FVC(r=-0.77、-0.78,P <0.05).Conclusion Detecting Tim-3 mRNA can effectively evaluate the occurrence and development of bronchial asthma,and it has an important reference value in clinical.
