【目的】探讨氧化低密度脂蛋白(ox‐LDL)对原代提取的小鼠骨髓间质干细胞(BMSCs)分泌胰岛素样生长因子(IGF‐1)、肝细胞生长因子(HGF )的影响。【方法】密度梯度离心法+贴壁培养法原代提取C57BL/6小鼠BMSCs ,检测细胞生长曲线和细胞表面抗原CD29、CD31、CD34、CD45;观察10μg/mL、20μg/mL、40μg/mL ox‐LDL作用48 h后BMSCs上清液中IGF‐1、HGF浓度的变化。【结果】刚分离的小鼠骨髓细胞呈圆形,经多次换液、传代后去除悬浮细胞,可见BMSCs贴壁生长,第三代细胞行流式细胞术检测表面抗原,显示CD29阳性,而 CD31、CD34、CD45阴性。ox‐LDL作用后BMSCs上清液中 IGF‐1、HGF浓度显著下降,且浓度越高下降越明显。【结论】ox‐LDL抑制体外培养的小鼠BMSCs分泌IGF‐1、HGF。
【目的】探討氧化低密度脂蛋白(ox‐LDL)對原代提取的小鼠骨髓間質榦細胞(BMSCs)分泌胰島素樣生長因子(IGF‐1)、肝細胞生長因子(HGF )的影響。【方法】密度梯度離心法+貼壁培養法原代提取C57BL/6小鼠BMSCs ,檢測細胞生長麯線和細胞錶麵抗原CD29、CD31、CD34、CD45;觀察10μg/mL、20μg/mL、40μg/mL ox‐LDL作用48 h後BMSCs上清液中IGF‐1、HGF濃度的變化。【結果】剛分離的小鼠骨髓細胞呈圓形,經多次換液、傳代後去除懸浮細胞,可見BMSCs貼壁生長,第三代細胞行流式細胞術檢測錶麵抗原,顯示CD29暘性,而 CD31、CD34、CD45陰性。ox‐LDL作用後BMSCs上清液中 IGF‐1、HGF濃度顯著下降,且濃度越高下降越明顯。【結論】ox‐LDL抑製體外培養的小鼠BMSCs分泌IGF‐1、HGF。
【목적】탐토양화저밀도지단백(ox‐LDL)대원대제취적소서골수간질간세포(BMSCs)분비이도소양생장인자(IGF‐1)、간세포생장인자(HGF )적영향。【방법】밀도제도리심법+첩벽배양법원대제취C57BL/6소서BMSCs ,검측세포생장곡선화세포표면항원CD29、CD31、CD34、CD45;관찰10μg/mL、20μg/mL、40μg/mL ox‐LDL작용48 h후BMSCs상청액중IGF‐1、HGF농도적변화。【결과】강분리적소서골수세포정원형,경다차환액、전대후거제현부세포,가견BMSCs첩벽생장,제삼대세포행류식세포술검측표면항원,현시CD29양성,이 CD31、CD34、CD45음성。ox‐LDL작용후BMSCs상청액중 IGF‐1、HGF농도현저하강,차농도월고하강월명현。【결론】ox‐LDL억제체외배양적소서BMSCs분비IGF‐1、HGF。
[Objective]To explore the impact of oxidized low density lipoprotein (ox‐LDL) on the secretion of insulin‐like growth factor 1 (IGF‐1) and hepatocyte growth factor (HGF) in murine bone marrow mesen‐chymal stem cells (MSCs) .[Methods]After primarily isolating and culturing bone marrow MSCs from C57BL/6 mice ,the cell growth curve was plotted .And flow cytometry was employed to determine cell surface anti‐gens of CD29 ,CD31 ,CD34 and CD45 .The impact of ox‐LDL on the secretion of IGF‐1 and HGF was as‐sessed in MSCs .[Results]The MSCs were CD29+ CD31‐CD34‐CD45‐cells and resembled the morphology of fibroblasts under inverted phase‐contrast microscope .MSCs could secret IGF‐1 and HGF and such effects were suppressed by different concentrations of ox‐LDL .[Conclusion]Ox‐ldl may suppress the secretion of IGF‐1 and HGF by MSCs .
