目的:建立瓜蒌皮中游离氨基酸类成分指纹图谱,并比较研究不同品种、产地瓜蒌皮氨基酸含量差异,为科学评价与有效控制瓜蒌皮质量提供依据。方法:采用异硫氰酸苯酯( PITC)为衍生试剂,色谱柱为Eclipse XDB C18(4.6mm ×250mm,5μm);流动相A:乙腈-水(4∶1),流动相B:醋酸钠-乙腈溶液(93∶7),梯度洗脱;流速:1mL/min;检测波长:254nm;柱温为30℃。结果:建立了瓜蒌皮中19种游离氨基酸类成分液相指纹图谱共有模式,并对不同产地瓜蒌皮药材进行了相似度比较和聚类分析。结论:本方法简便、快速、重复性好,可用于鉴别和区分不同来源的瓜蒌皮药材,为全面控制瓜蒌皮药材的质量提供依据。

목적:건립과루피중유리안기산류성분지문도보,병비교연구불동품충、산지과루피안기산함량차이,위과학평개여유효공제과루피질량제공의거。방법:채용이류청산분지( PITC)위연생시제,색보주위Eclipse XDB C18(4.6mm ×250mm,5μm);류동상A:을정-수(4∶1),류동상B:작산납-을정용액(93∶7),제도세탈;류속:1mL/min;검측파장:254nm;주온위30℃。결과:건립료과루피중19충유리안기산류성분액상지문도보공유모식,병대불동산지과루피약재진행료상사도비교화취류분석。결론:본방법간편、쾌속、중복성호,가용우감별화구분불동래원적과루피약재,위전면공제과루피약재적질량제공의거。
Objective:To set up fingerprints of amino acid of trichosanthes pericarpium from different varieties and habitats by HPLC,in order to establish a sensitive and specific method for controlling the quality of trichosanthes pericarpium. Methods:Phenyl isothiocyanate ( PITC) was used as a derivatization reagent.Chromatographic condition:Eclipse XDB C18 of HPLC column, (4.6mm ×250mm,5μm);mobile phase:gradient elution of acetonitrile-water (4∶1) (A) and sodium acetate solution-acetonitrile (93∶7) (B), detector wavelength:254nm;column temperature:30℃at the flow rate of 1mL/min.Results:The common modes of HPLC fingerprints of 19 kinds of amino acid in trichosanthes pericarpi-um were set up and the similar degrees to the crude drugs of different varieties and habitats were compared .Conclusion:The method is accurate, reliable and sensitive, which can be used as a quality control item for trichosanthes pericarpium collected from different sources and varieties.
