目的:探讨通天草提取物对阿尔茨海默氏病( Alzheimer’ s disease,AD)转基因小鼠的治疗作用。方法:每天2次使用灌胃器,将通天草提取物(0.3mL/10g· W)由小鼠口腔直接注入到小鼠胃中。通过Morris水迷宫和开场实验检测小鼠的学习记忆和探索能力。通过HE染色检测小鼠海马齿状回面积和细胞密度改变。结果:给予通天草提取物治疗28天后,在Morris水迷宫实验中,治疗组小鼠的逃避潜伏期明显缩短(P<0.01),经过平台次数明显多于对照组(P<0.01),停留在目标象限的时间明显延长(P<0.01);在开场实验中,治疗组小鼠穿经格子数和前肢抬起次数均显著多于对照组(P<0.01)。 HE染色结果显示,经通天草提取物治疗28天后,与对照组小鼠相比,治疗组小鼠海马齿状回面积明显增大(P<0.05),细胞密度显著增高(P<0.05)。结论:通天草提取物治疗可有效改善AD转基因小鼠学习记忆能力和空间探索能力。

목적:탐토통천초제취물대아이자해묵씨병( Alzheimer’ s disease,AD)전기인소서적치료작용。방법:매천2차사용관위기,장통천초제취물(0.3mL/10g· W)유소서구강직접주입도소서위중。통과Morris수미궁화개장실험검측소서적학습기억화탐색능력。통과HE염색검측소서해마치상회면적화세포밀도개변。결과:급여통천초제취물치료28천후,재Morris수미궁실험중,치료조소서적도피잠복기명현축단(P<0.01),경과평태차수명현다우대조조(P<0.01),정류재목표상한적시간명현연장(P<0.01);재개장실험중,치료조소서천경격자수화전지태기차수균현저다우대조조(P<0.01)。 HE염색결과현시,경통천초제취물치료28천후,여대조조소서상비,치료조소서해마치상회면적명현증대(P<0.05),세포밀도현저증고(P<0.05)。결론:통천초제취물치료가유효개선AD전기인소서학습기억능력화공간탐색능력。
Objective:To investigate the effect of waternut culm on Alzheimer's disease ( AD) transgenic mice.Methods:Waternut culm (0.3mL/10g· W) was injected from mice's mouth directly into stomach two times per day.The Morris water maze and open field test were used to investigate the learning and memory of mice.Histological test was performed to measure the alteration of the size and cell density of the dentate gyrus of mice.Results:28 days after treatment by wa-ternut culm extracts, compared with the mice in the control group, mice in the experiment group showed evidently short-ened escape latency(P<0.01), notably increased platform crossing(P<0.01) and prolonged time of staying in the target quadrant(P<0.01) in the Morris water maze test as well as significantly more active grid-passing (P<0.01) and forelimb-raising(P<0.01).HE staining showed that 28 days after treatment, mice in the experiment group pos-sessed larger dentate gyrus (P<0.05) and higher cell density in dentate gyrus (P<0.05) in contrast to the mice in the control group.Conclusion:Waternut culm is effective to imporove the learning and memory as well as space explora-tion of AD transgenic mice.
