目的:探讨 miR -542-5p 对卵巢癌细胞系 SKOV3的增殖、迁移、侵袭、凋亡等恶性生物学行为的影响。方法:应用 miR -542-5p mimics 和 mimics 阴性对照(mimics NC)分别转染卵巢癌细胞 SKOV3。采用MTT 实验、划痕试验、Transwell 实验和流式细胞术检测过表达 miR -542-5p 对 SKOV3细胞增殖、迁移、侵袭、凋亡的影响。结果:过表达 miR -542-5p 后,SKOV3细胞的增殖能力较阴性对照和正常 SKOV3细胞明显降低(P <0.05);Transwell 实验和划痕实验显示,过表达 miR -542-5p 后,细胞的迁移和侵袭能力较阴性对照组明显下降(P <0.05);流式细胞术结果显示,过表达 miR -542-5p 后,SKOV3细胞的凋亡率较阴性对照组升高,而对细胞周期无影响。结论:过表达 miR -542-5p 可以抑制卵巢癌细胞 SKOV3的体外增殖、迁移和侵袭能力,诱导细胞凋亡。提示 miR -542-5p 在卵巢癌恶性生物学进程中可能发挥重要作用。

목적:탐토 miR -542-5p 대란소암세포계 SKOV3적증식、천이、침습、조망등악성생물학행위적영향。방법:응용 miR -542-5p mimics 화 mimics 음성대조(mimics NC)분별전염란소암세포 SKOV3。채용MTT 실험、화흔시험、Transwell 실험화류식세포술검측과표체 miR -542-5p 대 SKOV3세포증식、천이、침습、조망적영향。결과:과표체 miR -542-5p 후,SKOV3세포적증식능력교음성대조화정상 SKOV3세포명현강저(P <0.05);Transwell 실험화화흔실험현시,과표체 miR -542-5p 후,세포적천이화침습능력교음성대조조명현하강(P <0.05);류식세포술결과현시,과표체 miR -542-5p 후,SKOV3세포적조망솔교음성대조조승고,이대세포주기무영향。결론:과표체 miR -542-5p 가이억제란소암세포 SKOV3적체외증식、천이화침습능력,유도세포조망。제시 miR -542-5p 재란소암악성생물학진정중가능발휘중요작용。
Objective:To investigate the effect of miR - 542 - 5p on the cell proliferation,migration,invasion and apoptosis of ovarian cancer SKOV3 cell. Methods:miR - 542 - 5p mimics and mimics negetive control(mimics NC) were transfected into SKOV3 cell lines. MTT assay,scratch test,Transwell assay and flow cytometry were performed to evaluate the effect of overexpression of miR - 542 - 5p on cell proliferation,migration,invasion and cel1 apoptosis of SKOV3 cell line. Results:The ability of proliferation of SKOV3 cell was significantly decreased compared with the ne-getive control group and parental SKOV3 cell group after overexpression of miR - 542 - 5p(P < 0. 05). Transwell and scratch tests indicated that after overexpression of miR - 542 - 5p,the invasion and migration ability of SKOV3 were significantly decreased than the negetive control group(P < 0. 05). Flow cytometry showed that the apoptosis rate of SKOV3 cell was increased after overexpression of miR - 542 - 5p compared with the negetive control group,but had no effect on cell cycle. Conclusion:The proliferation,migration and invasion of SKOV3 cell lines can be inhibited and the apoptosis rate can be increased by increasing the expression of miR - 542 - 5p. Our results indicate that miR -542 - 5p may play an important role in the malignant phenotypes of ovarian cancer.
