目的:探讨HBV X蛋白对阿霉素处理后的HepG2细胞凋亡影响及可能机制。方法用阿霉素处理HepG2、HepG2/GFP、HepG2/GFP-HBx细胞,分别用台盼兰染色和流式细胞仪检测各组细胞凋亡率;并通过RT-PCR和Western blot检测P53、PTEN的基因及蛋白表达。结果阿霉素处理36小时后, HepG2/GFP-HBx细胞凋亡率明显低于另外两组( P<0.01);HepG2/GFP-HBx细胞PTEN mRNA和蛋白水平均低于另外两组(P<0.01),而p53 mRNA和蛋白水平与另外两组无明显差别(P>0.05)。结论 HBV X蛋白能够抑制阿霉素诱导的HepG2细胞凋亡和PTEN的表达,HBV X蛋白可能通过影响p53-PTEN环路而抑制细胞凋亡。

목적:탐토HBV X단백대아매소처리후적HepG2세포조망영향급가능궤제。방법용아매소처리HepG2、HepG2/GFP、HepG2/GFP-HBx세포,분별용태반란염색화류식세포의검측각조세포조망솔;병통과RT-PCR화Western blot검측P53、PTEN적기인급단백표체。결과아매소처리36소시후, HepG2/GFP-HBx세포조망솔명현저우령외량조( P<0.01);HepG2/GFP-HBx세포PTEN mRNA화단백수평균저우령외량조(P<0.01),이p53 mRNA화단백수평여령외량조무명현차별(P>0.05)。결론 HBV X단백능구억제아매소유도적HepG2세포조망화PTEN적표체,HBV X단백가능통과영향p53-PTEN배로이억제세포조망。
Objective To investigate the effect of hepatitis B virus X protein in the Adriamycin-induced Apoptosis of HepG2 Cells and its possible mechanism.Methods HepG2, HepG2/GFP and HepG2/GFP-HBx cells were treated with adriamycin, and apoptotic cell death were examined by trypan blue exclusion and flow cytometry analysis.The transcription of p53, PTEN was analyzed by RT-PCR, and the expression of p53, PTEN was analyzed by western blot.Results After treated with adriamycin 36 hours,the apoptosis rate of HepG2/GFP-HBx was significantly lower than HepG2 cells and HepG2/GFP cells;the transcription and expression of PTEN in HepG2/GFP-HBx cells decreased, but there was no significant change in the transcription and expression of p53 in HepG2/GFP-HBx cells.Conclusions HBV X protein inhibited apoptosis of HepG2 cells induced by adriamycin and PTEN expression.The inhibitory effect of HBV X protein on the cell apoptosis may be related to the inhibition of p53-PTEN pathway.
