目的探讨标本放置时间和温度对胰岛素和C肽结果的影响,以期建立实验室标本存放标准。方法30例标本采用化学发光的方法检测其胰岛素和C肽水平,并置于室温(21℃)、4℃和-20℃,测定不同保存时间的胰岛素和C肽水平。结果血清置-20℃温度24h和1w,4℃温度4h与即时检测结果相比无统计学意义(>0.05);血清置室温温度(21℃)1h,置4℃温度12h与即时检测结果相比均有统计学意义(<0.05);标本经室温保存2、3h,4℃保存72h与-20℃保存4w(672h)后,胰岛素和C肽水平显著下降(<0.01)。结论标本冷冻是准确检测血清胰岛素和C肽的金标准,而室温下保存可能降低分析物的检测水平。血浆样品储存在4℃,应在静脉穿刺后4h内进行分析。

Objective To investigate the impact of storage time and temperature on test results of insulin and C-peptide, and to develop laboratory Standards for Samples Storage . Methods The serum insulin and C peptide levels in 30 blood samples were determined by chemiluminescence immunoassay. Samples were stored at room temperature, 4℃ and -20℃, and then analyzed at dif erent times. Results Compared with Point-Of-Care Testing(POCT), there was no substantial change in insulin and C peptide levels when blood samples stored at-20℃for 24 or 168 hours and 4℃for 4 hours( >0.05).At room temperature for 1 hour and 4℃for 12 hours,There was significantly dif erence of insulin and C peptide levels when compared to POCT ( <0.05). Significant changes were also observed for insulin and C peptide (decrease at 2 and 3 hours, room temperature;72hours, 4℃;672hours,-20℃)( <0.01).Conclusion Our study confirmed that sample freezing is the gold standard for accurate determination of serum insulin and C peptide, that storage at room temperature may decrease the analyte levels. Plasma samples should be stored at 4oC and analysed within 4 hours of venepuncture.
