目的:探讨TLR2在临床口腔鳞癌患者中的表达情况,以及TLR2在口腔鳞癌发生和发展过程中的作用,进而为临床口腔鳞癌的干预和治疗寻找新的治疗途径。方法无菌提取24例口腔鳞癌患者、24例良性肿瘤患者及24例对照组外周血单核细胞,利用实时定量PCR及Western blot方法检测TLR2的表达情况,同时通过免疫组化技术来检测癌组织中的TLR2表达水平的差异;通过TLR2激动剂(弗氏完全佐剂)及弗氏不完全佐剂(阴性对照组)作用口腔鳞癌成瘤小鼠,观察其瘤体的生长及荷瘤小鼠的存活情况。结果口腔鳞癌患者外周血单核细胞中TLR2的表达较之对照组明显增加(P<0.05),同时TLR2表达水平在口腔鳞癌组织中也明显增强。注射弗氏佐剂组的小鼠瘤体的体质量为(1.42±0.43)g,较之空白对照组[(2.47±0.69)g]及阴性对照组(弗氏不完全佐剂组)[(2.94±0.87)g]明显降低,同时小鼠的生存期也明显延长(P<0.05)。结论口腔鳞癌患者外周血单核细胞及癌组织中TLR2的表达明显升高,同时TLR2的高表达具有明显的肿瘤抑制作用。

목적:탐토TLR2재림상구강린암환자중적표체정황,이급TLR2재구강린암발생화발전과정중적작용,진이위림상구강린암적간예화치료심조신적치료도경。방법무균제취24례구강린암환자、24례량성종류환자급24례대조조외주혈단핵세포,이용실시정량PCR급Western blot방법검측TLR2적표체정황,동시통과면역조화기술래검측암조직중적TLR2표체수평적차이;통과TLR2격동제(불씨완전좌제)급불씨불완전좌제(음성대조조)작용구강린암성류소서,관찰기류체적생장급하류소서적존활정황。결과구강린암환자외주혈단핵세포중TLR2적표체교지대조조명현증가(P<0.05),동시TLR2표체수평재구강린암조직중야명현증강。주사불씨좌제조적소서류체적체질량위(1.42±0.43)g,교지공백대조조[(2.47±0.69)g]급음성대조조(불씨불완전좌제조)[(2.94±0.87)g]명현강저,동시소서적생존기야명현연장(P<0.05)。결론구강린암환자외주혈단핵세포급암조직중TLR2적표체명현승고,동시TLR2적고표체구유명현적종류억제작용。
Objective To explore the expression of Toll-like receptor 2(TLR2)in oral squamous cell carcinoma(OSCC) and its role in the development of tumor. Methods The expression of TLR2 was detected in peripheral monocyte of 24 cases of OSCC, 24 cases of benign tumor and 24 normal controls by RT-PCR and Western blot. And the TLR2 expression in tumor tissues was detected by immuno-histochemical method. Freund’ s complete adjuvant( CFA) and incomplete adjuvant( IFA) were injected into mice to observe the sur-vival time of tumor-bearing mice and the tumor growth. Results The expression of TLR2 mRNA in peripheral monocyte was higher in OSCC than those in normal controls or in benign tumor(P<0. 05). And the TLR2 protein levels were also higher in tumor tissues of OSCC than in normal controls or in benign tumor(P<0. 05). The tumor weight was(1. 42 ± 0. 43)g in mice injected with CFA,(2. 94 ± 0. 87)g in mice injected with incomplete adjuvant(IFA) and (2. 47 ± 0. 69)g in blank control mice. The survival time of the tumor-bearing mice in CFA group was significantly prolonged compared with IFA group and blank group(P<0. 05). Conclusion TLR2 expression increases in peripheral monocyte and tumor tissues of OSCC patients,and TLR2 high expression significantly inhibit the pro-liferation of human tongue squamous cancer cells.
