目的:采用动态优化方法,优化何首乌的提取工艺。方法:以2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷、大黄素-8-O-β-D-葡萄糖及大黄素甲醚-8-O-β-D-葡萄糖含量为指标,结合指纹图谱相似度分析,比较何首乌正交优化提取工艺和动态优化提取工艺所得提取物的一致性。结果动态优化提取工艺为10倍水提3次,第一次90min,第二次70min,第三次50min。两种工艺所得提取物中指标成分含量及浸膏得率的RSD均<5%,指纹图谱相似度为100%,表明两种提取物基本一致;动态优化提取工艺比正交优化提取工艺节约61%的时间,能大幅降低成本。结论动态优化方法在何首乌的提取工艺上是可行的,并能节约生产成本。

목적:채용동태우화방법,우화하수오적제취공예。방법:이2,3,5,4’-사간기이분을희-2-O-β-D-포도당감、대황소-8-O-β-D-포도당급대황소갑미-8-O-β-D-포도당함량위지표,결합지문도보상사도분석,비교하수오정교우화제취공예화동태우화제취공예소득제취물적일치성。결과동태우화제취공예위10배수제3차,제일차90min,제이차70min,제삼차50min。량충공예소득제취물중지표성분함량급침고득솔적RSD균<5%,지문도보상사도위100%,표명량충제취물기본일치;동태우화제취공예비정교우화제취공예절약61%적시간,능대폭강저성본。결론동태우화방법재하수오적제취공예상시가행적,병능절약생산성본。
OBJECTIVE To optimize the extraction method of Polygonum multiflorum Thumb.by applying dy-namic optimization extraction method ( DOEM).METHODS Stilbene glucoside ,emodin-8-O-β-D-glucose and emo-din methyl ether-8-O-β-D-glucose were regarded as indexes ,two extraction processes ,orthogonal design and DOEM , were both employed to optimize extraction of Polygonum multiflorum thumb.,and then were compared by concents of marker components ,extract yield and similarity evaluation of fingerprint .RESULTS The dynamic optimization ex-traction process was extracting for 3 times with 10-folds of water,and extracting time was 90,70,50 minutes in turn.Relative standard deviations of concentration of marker components and total solids yield in these extraction processes were less than 5%with a 100%of similarity in fingerprint .The DOEM process could save 61%of the ex-traction time and greatly reduce the cost , which was superior to the orthogonal design optimization process.CONCLUSION Dynamic optimization extraction method on the extraction process of Polygonum multiflo-rum Thumb.is feasible ,and can save the cost of production .
