目的:探讨氧化巴西苏木素对BALB/c小鼠坐骨神经损伤后相应脊髓节段中NF-κB表达的影响。方法健康成年雄性BALB/c小鼠160只,行单侧坐骨神经离断缝合术后,随机分为空白对照组,高剂量组,中剂量组和低剂量组四组,同期给药。于给药后存活不同时间点获取术侧L4—L6脊髓节段,应用Westen-Blot法检测其中NF-κB的表达变化。于术后第1,2,4,8周行电生理检测。结果 Westen-Blot结果显示高剂量和中剂量组12 h,24 h,3 d,5 d,1周NF-κB 表达量明显低于低剂量组和空白对照组。2周,4周,8周各组比较无明显差异。电生理检测各剂量组与对照组间均存在显著的差异。结论氧化巴西苏木素对坐骨神经损伤后脊髓相应节段细胞中NF-κB活化起到抑制作用,可促进神经再生。

목적:탐토양화파서소목소대BALB/c소서좌골신경손상후상응척수절단중NF-κB표체적영향。방법건강성년웅성BALB/c소서160지,행단측좌골신경리단봉합술후,수궤분위공백대조조,고제량조,중제량조화저제량조사조,동기급약。우급약후존활불동시간점획취술측L4—L6척수절단,응용Westen-Blot법검측기중NF-κB적표체변화。우술후제1,2,4,8주행전생리검측。결과 Westen-Blot결과현시고제량화중제량조12 h,24 h,3 d,5 d,1주NF-κB 표체량명현저우저제량조화공백대조조。2주,4주,8주각조비교무명현차이。전생리검측각제량조여대조조간균존재현저적차이。결론양화파서소목소대좌골신경손상후척수상응절단세포중NF-κB활화기도억제작용,가촉진신경재생。
Objective To investigate the effect of Brazilein on the expression of NF-κB in spinal motor neurons in mice after sciatic nerve injury. Methods 160 adult male mice were randomly divided into 4 groups after sciatic nerve injury: blank control group, the high dose group, the middle dose group and the low dose group, simultaneous treatment respectively. The L4~L6 spinal segments of the injured side were obtained for Westen-Blot. Electrophysiological examination were carried out at 1,2,4,8 weeks after injury. Results The results of Westen-Blot showed at 12 h, 3 d, 5 d, 1 w, the expression of NF-κB in the high dose group and the middle dose group were significantly lower than those in the low dose group and blank control group. At 2 w, 4 w, 8 w there were no significant differences. Electrophysiological examination showed significant differences between treatment group and blank control group. Conclusion Brazilein can enhance regeneration of peripheral nerve by inhibiting the activation of NF-κB in spinal motor neurons in mice after sciatic nerve injury.
