为优化生防菌哈茨木霉FJA T‐9040固体发酵条件,结合初始含水量、初始接种量及培养温度3个单因素,Box‐Behnken设计和响应面分析法进行研究。以产孢量为指标,单因素试验确定初始含水量、初始接种量及培养温度分别为50%、5%、28℃。响应面分析结果表明:初始含水量和温度与产孢量有显著相关性( P<0.05),通过求解回归方程可得,当初始含水量为53%,初始接种量为7%,培养温度29℃时,产孢量达到4.04×109个· g -1,与实际值3.86×109个· g -1相近。

위우화생방균합자목매FJA T‐9040고체발효조건,결합초시함수량、초시접충량급배양온도3개단인소,Box‐Behnken설계화향응면분석법진행연구。이산포량위지표,단인소시험학정초시함수량、초시접충량급배양온도분별위50%、5%、28℃。향응면분석결과표명:초시함수량화온도여산포량유현저상관성( P<0.05),통과구해회귀방정가득,당초시함수량위53%,초시접충량위7%,배양온도29℃시,산포량체도4.04×109개· g -1,여실제치3.86×109개· g -1상근。
Optimization of solid‐state fermentation condition for the biocontrol strain of T richoderma harz ianum FJAT‐9040 using single factor analysis ,Box‐Behnken design.and response surface methodology (RSM ) . The constitute of fermentation condition initial moisture ,initial inoculation amount ,culture temperature were determined by single factor experiment ,then the optimal condition was optimized using Box‐Behnken design and RSM. The result of single factor showed that the initial moisture ,initial inoculation amount ,culture temperature were 50% , 5% ,28℃ ,respectively. The results showed that initial moisture and culture temperature were significant for the biomass of strain of T.harz ianum based on the response surface methodology analysis ( P< 0.05 ) .T he optimum fermentation conditions were as followed :initial moisture 53% ,initial inoculation amount 7% ,culture temperature 29℃ , the average sporulation yield reached to 4.04 × 109 cfu · g-1 , which was closed to the actual 3.86 × 109 cfu · g-1 .
