目的:探讨新鲜冰冻血浆对脑损伤合并失血性休克大鼠的治疗作用。方法取25只健康雌性 SD 大鼠,采用Aghaloo 法制备新鲜冰冻血浆。将108只健康成年 SD 大鼠随机分为3组:sham 组18只仅开放颅骨骨窗;vehicle 组与FFP 组(各45只)采用 Feeney 改良法制作脑损伤模型与 Wiggers 改良法制作失血性休克模型,60 min 后 vehicle 组给予3倍失血量的生理盐水复苏,FFP 组给予同等失血量 FFP 进行复苏。伊文氏蓝(EB)染色测定血脑屏障通透性,干湿重法测量脑组织含水量,改良神经功能评分(mNSS)评价大鼠神经功能障碍改善情况,7 d 时脑组织切片尼氏染色观察神经元细胞生存情况。结果1、3、7 d 时 FFP 组脑水肿程度比 vehicle 组轻(P <0.05);12、24、72 h 时各组脑组织 EB 含量差异有统计学意义(P <0.05),12、24、72 h 时 FFP 组比 vehicle 组 EB 含量低(P <0.05);3、7 d 时 FFP 组 mNSS 神经功能评分比 vehicle 组低(P <0.05)。结论早期采用新鲜冰冻血浆对脑损伤合并失血性休克大鼠进行治疗可以减轻脑水肿,降低血脑屏障通透性,改善神经功能。

목적:탐토신선빙동혈장대뇌손상합병실혈성휴극대서적치료작용。방법취25지건강자성 SD 대서,채용Aghaloo 법제비신선빙동혈장。장108지건강성년 SD 대서수궤분위3조:sham 조18지부개방로골골창;vehicle 조여FFP 조(각45지)채용 Feeney 개량법제작뇌손상모형여 Wiggers 개량법제작실혈성휴극모형,60 min 후 vehicle 조급여3배실혈량적생리염수복소,FFP 조급여동등실혈량 FFP 진행복소。이문씨람(EB)염색측정혈뇌병장통투성,간습중법측량뇌조직함수량,개량신경공능평분(mNSS)평개대서신경공능장애개선정황,7 d 시뇌조직절편니씨염색관찰신경원세포생존정황。결과1、3、7 d 시 FFP 조뇌수종정도비 vehicle 조경(P <0.05);12、24、72 h 시각조뇌조직 EB 함량차이유통계학의의(P <0.05),12、24、72 h 시 FFP 조비 vehicle 조 EB 함량저(P <0.05);3、7 d 시 FFP 조 mNSS 신경공능평분비 vehicle 조저(P <0.05)。결론조기채용신선빙동혈장대뇌손상합병실혈성휴극대서진행치료가이감경뇌수종,강저혈뇌병장통투성,개선신경공능。
Objective To investigate the efficacy of fresh -frozen plasma (FFP)on brain injury accompanied by hemorrhagic shock in rats.Methods 25 healthy SD rats were selected and FFP was prepared.108 healthy adult SD rats were randomly divided into three groups.The sham group (18 cases)only opened skull bone window,vehicle group and FFP group (45 cases in each) used Feeney improved method and Wiggers improved method to make rat brain injury and hemorrhagic shock model.After 60 mi-nutes,the vehicle group were given three times the amount of blood loss saline,FFP group were given the same amount of blood loss FFP.Blood -brain barrier permeability were measured by EB staining,cerebral edema were measured by dry -wet weight method.The improvement of neurological disorders were measured by mNSS score,and brain neuronal cell survival situation were deserved by Nissl staining on the 7th day.Results On the 1st,3rd,7th day,brain edema of FFP group was lower than vehicle group (P <0.05).In 12,24,72 h,EB content of FFP group was lower than the vehicle group (P <0.05),mNSS neurological scores of FFP group were lower than the vehicle group on the 3rd,7th day.Conclusion FFP could relieve brain edema,reduce the permeability of blood -brain barrier,improve nerve function in the treatment of brain injury and hemorrhagic shock resuscitation in rats.
