目的观察SW480结肠癌细胞上皮-间充质转化(EMT)后β1整合素表达变化,探讨其对结肠癌细胞体外侵袭力的影响.方法用10 μg/L转化生长因子-β1(TGF-β1)处理72 h后检测SW480结肠癌细胞发生EMT.实时定量聚合酶链反应(Real-time PCR)及Western blot方法分别检测转化前后癌细胞β1整合素mRNA和蛋白的表达;体外侵袭实验检测转化前后SW480细胞的体外侵袭能力.结果 EMT后结肠癌细胞β1整合素mRNA表达较转化前明显增强(t=3.764,P＜0.05);EMT后结肠癌细胞β1整合素蛋白表达(0.430±0.021)较转化前(0.172 ±0.034)明显增强(P＜ 0.05);Transwell细胞侵袭实验显示EMT后结肠癌细胞的穿膜细胞数[(137±9)个]较转化前[(36±4)个]明显增多(P＜0.05).结论结肠癌细胞EMT后β1整合素表达升高,其体外侵袭力增强.
목적 관찰SW480결장암세포상피-간충질전화(EMT)후β1정합소표체변화,탐토기대결장암세포체외침습력적영향.방법 용10 μg/L전화생장인자-β1(TGF-β1)처리72 h후검측SW480결장암세포발생EMT.실시정량취합매련반응(Real-time PCR)급Western blot방법분별검측전화전후암세포β1정합소mRNA화단백적표체;체외침습실험검측전화전후SW480세포적체외침습능력.결과 EMT후결장암세포β1정합소mRNA표체교전화전명현증강(t=3.764,P＜0.05);EMT후결장암세포β1정합소단백표체(0.430±0.021)교전화전(0.172 ±0.034)명현증강(P＜ 0.05);Transwell세포침습실험현시EMT후결장암세포적천막세포수[(137±9)개]교전화전[(36±4)개]명현증다(P＜0.05).결론 결장암세포EMT후β1정합소표체승고,기체외침습력증강.
Objective To investigate the expression of intergin-β1 of human colon carcinoma cell line SW480 after epithelial-mesenchymal transition (EMT) and effect of invasive ability on the human colon carcinoma in vitro.Methods The epithelial-mesenchymal transition of SW480 cells was introduced by transforming growth factor-β1 (TGF-β1,10 μg/L) for 72 h.The expression of intergin-β1 in SW480 cells was detected by real-time quantitative polymerase chain reaction (Real-time PCR) and Western blotting.The migration of SW480 cells in vitro was assayed by Transwell.Results In contrast with that before EMT,the expression of intergin-β1 mRNA in SW480 cells after EMT was increased obviously (t =3.764,P ＜ 0.05).As compared with that before EMT (0.172 ± 0.034),the expression of intergin-β1 protein in SW480 cells after EMT (0.430 ±0.021) was increased obviously (P ＜0.05).The migration of SW480 cells after EMT (137 ± 9) was significantly increased (P ＜ 0.05) as compared with that before EMT (36 ±4).Conclusion The expression of intergin-β1 and the migration of human colon carcinoma cells in vitro can be elevated after EMT.
