目的 观察血小板膜糖蛋白CD61,CD62P在脓毒症小鼠炎性反应中的调控作用.方法 采用盲肠结扎穿孔术(CLP)制作小鼠脓毒症模型;将60只BALB/c雄性小鼠随机分为阴性对照组、假手术组、CLP组、CLP+血小板膜糖蛋白受体拮抗剂替罗非班组(替罗非班组).检测处理后24 h外周血血小板CD61、CD62P和血清肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-6的表达.结果 CLP组血小板计数为(167.0 ±31.4)×106/L较阴性对照组[(289.7 ±45.9)×106/L]、假手术组[(267.7±25.4)×106/L]、替罗非班组[(217.0 ±29.7) ×106/L]明显降低(P<0.01);CLP组CD61、CD62P[(91.22 ±2.96)%、(7.87±2.90)%]及TNF-α、IL-6[(723.51±96.65)、(757.35±131.48) ng/L]表达较对照组明显升高(P<0.01),替罗非班组CD61、CD62P[(67.50±8.67)%、(3.53±0.78)%]及TNF-α、IL-6[(426.38±16.69)、(482.32±56.73)ng/L]表达较CLP组降低(P<0.01);脓毒症小鼠血小板膜糖蛋白CD61、CD62P表达与炎性因子的表达呈正相关.结论 血小板膜糖蛋白CD61、CD62P可能通过调控炎性因子表达参与脓毒症炎性反应的病理生理过程.

목적 관찰혈소판막당단백CD61,CD62P재농독증소서염성반응중적조공작용.방법 채용맹장결찰천공술(CLP)제작소서농독증모형;장60지BALB/c웅성소서수궤분위음성대조조、가수술조、CLP조、CLP+혈소판막당단백수체길항제체라비반조(체라비반조).검측처리후24 h외주혈혈소판CD61、CD62P화혈청종류배사인자-α(TNF-α)、백세포개소(IL)-6적표체.결과 CLP조혈소판계수위(167.0 ±31.4)×106/L교음성대조조[(289.7 ±45.9)×106/L]、가수술조[(267.7±25.4)×106/L]、체라비반조[(217.0 ±29.7) ×106/L]명현강저(P<0.01);CLP조CD61、CD62P[(91.22 ±2.96)%、(7.87±2.90)%]급TNF-α、IL-6[(723.51±96.65)、(757.35±131.48) ng/L]표체교대조조명현승고(P<0.01),체라비반조CD61、CD62P[(67.50±8.67)%、(3.53±0.78)%]급TNF-α、IL-6[(426.38±16.69)、(482.32±56.73)ng/L]표체교CLP조강저(P<0.01);농독증소서혈소판막당단백CD61、CD62P표체여염성인자적표체정정상관.결론 혈소판막당단백CD61、CD62P가능통과조공염성인자표체삼여농독증염성반응적병리생리과정.
Objective To observe the regulation of platelet membrane glycoprotein CD61,CD62P in septic inflammation.Methods Make the model of sepsis by cecal ligation and puncture (CLP).Sixty male BALB/c mice were randomly divided into 4 groups (negative control,sham operation,CLP,CLP + Tirofiban).The platelet counts,expressions of CD61 and CD62P in peripheral platelets,levels of tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 in serum were measured at 24 h after operation.Resuits Compared with those of negative control group [(289.7 ± 45.9) × 106/L],sham operation group [(267.7 ±25.4) × 106/L] and CLP + Tirofiban group [(217.0 ± 29.7) × 106/L],the platelet count was lower in mice with CLP [(167.0 ±31.4) × 106/L,P <0.01].The expressions of CD61 [(91.22 ± 2.96)%],CD62P [(7.87 ±2.90)%],TNF-α [(723.51 ±96.65) ng/L] and IL-6 [(757.35 ± 131.48) ng/L] in mice with CLP increased significantly (P < 0.01).However,the expressions of CD61 [(67.50±8.67)%],CD62P [(3.53 ±0.78)%],TNF-α [(426.38 ± 16.69) ng/L] and IL-6 [(482.32 ±56.73) ng/L] in mice with CLP + Tirofiban were all lower than that of CLP group (P< 0.01).There is positive correlation between the expression of platelet membrane glycoprotein and the expression of inflammatory factors in mice with CLP.Conclusion Platelet activation is closely related to the inflammatory response of sepsis.Down-regulation of platelet membrane glycoprotein can significantly attenuate the septic inflammation.
