目的 探讨WEE1在结肠癌组织中的表达及其临床意义.方法 实时定量聚合酶链反应(Real-time PCR)法检测79例结肠癌组织中WEE1的表达;Kendall' tau法进行相关性分析;Kaplan-Meier法绘制生存曲线,Log-rank检验分析;Cox比例风险回归模型分析独立预后影响因素.结果 WEE1的表达与结肠癌患者年龄、性别、肿瘤分化程度、TNM分期等因素无显著相关,但与结肠癌术后复发显著相关(r =0.297,P<0.01),WEE1表达阴性的患者具有较高的复发率(28.85%比3.70%,P<0.01);Cox比例风险回归模型分析显示WEE1表达水平是结肠癌的独立预后影响因素(P<0.05),与WEE1表达阳性的患者比较,WEE1表达阴性的患者具有较短的中位生存期(45个月比85个月,P <0.05).结论 WEE1参与结肠癌的发生发展,是结肠癌独立的预后影响因素.

Objective To investigated the clinico-pathological and prognostic value of WEE1 expression in colon cancer.Methods Real-time quantitative polymerase chain reaction (Real-time PCR) was employed to detect the expression level of WEE1 in colon cancer tissues of 79 patients.Correlation between variables was analyzed by Kendall' tau method.Survival curves were plotted using Kaplan-Meier method with Log-rank test.Cox proportional hazard model was used to evaluate the independent prognostic factors.Results There was no significant correlation between WEE1 expression and gender,age,tumor differentiation,TNM stage,et al.Patients lacking of WEE1 expression had a higher recurrence rate (28.85% vs.3.70%,P <0.01) and a poorer prognosis (P < 0.01).Cox regression analysis indicated that WEE 1 expression was a significant prognostic factor (P < 0.05).Conclusion WEE1 expression was a significant independent prognostic factor of colon cancer patients.
