目的 观察右旋美托咪啶对内毒素休克大鼠海马炎性反应和认知功能损伤的影响.方法 60只雄性SD大鼠随机分为4组:生理盐水对照组、内毒素休克组、高剂量右旋美托咪啶+内毒素血症组(High-dose Dex组)和低剂量右旋美托咪啶+内毒素血症组(Low-dose Dex组).酶联免疫吸附试验(ELISA)法检测海马白细胞介素(IL)-6及肿瘤坏死因子-α(TNF-α)表达水平.Western blot法检测海马中凋亡标志物活化半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3的表达水平.Morris水迷宫法测定认知功能.结果 每小时2.0μg/kg和0.5μg/kg右旋美托咪啶分别使内毒素休克大鼠海马IL-6表达下降67.1%和36.5% (P<0.05),TNF-α表达下降56.7%和53.1%(P<0.05),活化Caspase-3水平下降32.5%和38.5% (P <0.05),并缓解内毒素休克大鼠的认知功能障碍(P<0.05).结论 右旋美托咪啶可缓解内毒素休克大鼠海马炎性反应并减轻内毒素休克导致的认知功能损伤.

Objective To investigate the effects of dexmedetomidine (Dex) on hippocampal inflammatory responses and cognitive impairment in rats with endotoxin-induced shock.Methods Sixty male SD rats were randomly divided into 4 groups including saline control group,high-dose Dex group and low-dose Dex group.Hippocampal protein levels of interleukin (IL)-6 and tumor necrosis factor-α (TNF-α) were detected by enzyme linked immunosorbent assay (ELISA).Hippocampal active Caspase -3 protein expression levels were examined by Western blotting.Results As compared with saline control group,2.0 μg/(kg· h) and 0.5 μg/(kg·h) Dex respectively decreased hippocampal IL-6 protein levels by 67.1% and 36.5% (P < 0.05),and decreased hippocampal TNF-α protein levels by 56.7% and 53.1% (P < 0.05),reduced hippocampal active Caspase-3 levels by 32.5% and 38.5% (P < 0.05) and then attenuated LPS-induced cognitive impairment (P < 0.05).Conclusion Dex can reduce hippocampal inflammatory responses and attenuate cognitive impairment caused by endotoxin-induced shock.
