Haptoglobin%Papillary thyroid carcinoma
目的 探讨甲状腺乳头状癌组织中触珠蛋白(Hp)的表达及意义.方法 采用反转录-聚合酶链反应(RT-PCR)、Western blot技术检测甲状腺乳头状癌组织、结节性甲状腺肿及正常甲状腺组织中Hp mRNA和蛋白的表达.结果 甲状腺乳头状癌组织中Hp mRNA的相对表达量(0.913 ±0.102)明显高于结节性甲状腺肿(0.357±0.009)和正常甲状腺组织(0.296 ±0.008).Hp蛋白在甲状腺乳头状癌组织中的相对表达量为0.892 ±0.122,显著高于结节性甲状腺肿(0.211 ±0.051)和正常甲状腺组织(0.197±0.046,P<0.05),而后两者之间差异无统计学意义(P>0.05).随着TNM分期的增加,Hp mRNA和蛋白表达水平有升高的趋势,但各分期间的表达差异无统计学意义(P>0.05).结论 Hp在甲状腺乳头状癌的发生发展中发挥重要作用.

Objective To explore the expression and significance of haptoglobin (Hp) in papillary thyroid carcinoma.Methods Reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting were applied to detect the expression of Hp in 112 cases of papillary thyroid carcinoma,67 cases of benign thyroid node and 50 cases of normal thyroid tissues.Results The relative expression value of Hp mRNA was (0.913 ±0.102) in papillary thyroid carcinoma tissues,which were over-expressed comparing with benign thyroid node (0.357 ± 0.009) and normal thyroid tissue (0.296 ± 0.008) groups (P < 0.05).The relative expression value of Hp protein was (0.892 ± 0.122) in papillary thyroid carcinoma tissues,which were up-regulated comparing with benign thyroid node (0.211 ±0.051) and normal thyroid tissue (0.197 ± 0.046) groups (P < 0.05).However,there was no statistical significance between benign thyroid node and normal thyroid tissues (P > 0.05).In addition,although the level of Hp mRNA increased with the higher clinical stages,there were no statistical significance (P > 0.05).Condusion Our results suggested that Hp might play a key role in papillary thyroid carcinoma occurrence.Moreover,findings in the present study also provided evidence for the diagnostic and therapeutic strategies in the treatment of papillary thyroid carcinoma.
