目的探讨沉默信息调节因子2相关酶1(SIRT1)在人类甲状腺乳头状癌组织中的表达及其意义.方法利用实时荧光定量聚合酶链反应(FQ-PCR)、Western blot法检测40例甲状腺乳头状癌组织及相应的40例癌旁正常组织中SIRT1的mRNA、蛋白表达水平并进行统计学分析.结果 SIRT1 mRNA及蛋白表达在甲状腺乳头状癌组织中的表达量(分别为2.27±0.12、0.843±0.101)显著高于癌旁正常甲状腺组织(0.32±0.06、0.045±0.009,P＜0.01).SIRT1 mRNA及蛋白表达在甲状腺乳头状癌淋巴结转移组(分别为3.52±0.17、1.327±0.042)高于无淋巴结转移组(1.25±0.09、0.447±0.024),差异有统计学意义(P＜0.05).SIRT1 mRNA及蛋白表达与患者的性别(男2.37±0.19、0.903±0.112,女2.22±0.11、0.811±0.088)、年龄(＜45岁组2.11±0.14、0.805±0.107,≥45岁组2.54±0.16、0.906±0.117)、肿瘤大小(＜3 cm组2.21±0.15、0.810±0.043,≥3 cm组2.36±0.18、0.891±0.110)、包膜侵犯(有侵犯组2.26±0.24、0.840±0.077,无侵犯组2.27±0.13、0.845±0.089)及TNM分期(Ⅱ～Ⅱ期2.08±0.13、0.810±0.041,Ⅲ～Ⅳ期2.71±0.21、0.920±0.083)均无明显相关(P＞0.05).结论 SIRT1 mRNA及其蛋白的异常表达可能与甲状腺乳头状癌的发生发展和淋巴结转移有关.

Objective To investigate the expression and clinical significance of silent mating type information regulation 1 (SIRT1) in human papillary thyroid carcinoma.Methods Real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) and Western blotting were performed to detect the expression and distribution of mRNA and protein of SIRT1 in papillary thyroid carcinoma tissues (40 cases) and adjacent normal tissues (40 cases).Statistics was used to analyze the datas.Results The expression of SIRT1 mRNA and protein in papillary thyroid carcinoma (2.27 ±0.12,and0.843 ±0.101) was significantly higher than that in adjacent normal tissues (0.32 ± 0.06,and 0.045 ± 0.009,P ＜ 0.01).The expression of SIRT1 mRNA and protein was higher in the group with lymph node metastasis (3.52 ±0.17,and 1.327 ±0.042) than in the group without lymph node metastasis (1.25 ±0.09,and 0.447 ±0.024,P ＜ 0.05).The expression of SIRT1 mRNA and protein in papillary thyroid carcinoma was not related to patients' gender (male:2.37 ±0.19,and 0.903 ±0.112,and female:2.22 ±0.11,and 0.811 ± 0.088),age (＜45 years old:2.11 ±0.14,and 0.805 ±0.107,and ≥45 years old:2.54 ±0.16,and 0.906 ±0.117),tumor size (＜3 cm:2.21 ±0.15,and 0.810 ±0.043,and ≥3 cm:2.36 ±0.18,and 0.891 ± 0.110),capsule invasion (positive:2.26 ± 0.24,and 0.840 ± 0.077,and negative:2.27 ± 0.13,and 0.845 ±0.089) or TNM stage (Ⅰ-Ⅱ stage:2.08 ±0.13,and 0.810 ±0.041,and Ⅲ-Ⅳ stage:2.71 ± 0.21,and 0.920 ± 0.083,P ＞ 0.05).Conclusion The abnormal expression of SIRT1 may be associated with the oncogenesis and progress of papillary thyroid carcinoma and lymph node metastasis.
