目的 观察坐骨神经慢性压榨损伤(CCI)模型大鼠脊髓灰质后角组蛋白乙酰转移酶P300与CREB结合蛋白(CBP)表达的变化及其细胞分布.方法 将雄性SD大鼠48只随机分为空白组(12只)、假手术组(12只)、CCI造模组(24只).建模成功后,应用免疫组织化学法及免疫荧光法观察P300及CBP的表达及定位.结果 与空白组和假手术组比较,CCI模型组大鼠脊髓灰质后角细胞核内P300/CBP表达量上升(P>0.05),并伴有机械痛与热痛行为学特征,且于造模后14d达到峰值(P300数值为478.35±22.61,CPB为891.55±32.19),采用免疫荧光法观察与其共定位的细胞为神经元细胞.结论 P300/CBP与神经病理性痛的发生有关,且在疾病发展过程中仅分布于脊髓灰质后角的神经元细胞内.

Objective To investigate the distribution and dynamic changes of P300/CREB binding protein (CBP) expression in the spinal cord in the rat chronic constrictive injury (CCI) model.Methods Thermal withdrawal latency and mechanical withdrawal threshold were used to evaluate the CCI surgery.The number of IR cells was showed increased P300 and CBP immunoreactivity (IR) compared with normal control and sham-operated rats.Cell-type determination by immunofluorescence at 14 days.Results P300 and CBP expressed predominantly in nuclei in the gray matter of rat spinal cord.Rats undergoing CCI surgery showed increased IR of P300 and CBP compared with normal control and sham-operated rats(P >0.05).The number of IR cells reached the peak at 14 days following CCI compared with those on 3,7,and 21 days,accompanied with significant behavioral changes of neuropathic pain.Cell-type determination by immunofluorescence at 14 days following CCI revealed that P300 and CBP expressed in neurons.Conclusion These results suggest that P300 and CBP are probably involved in the maintenance of neuropathic pain on spinal cord level.Furthermore,P300 and CBP may serve as a sensor only in neurons in the adult rat spinal cord.
