目的:探讨左归丸和巴氯芬联用对新生脑瘫大鼠运动协调能力和认知能力的影响,并研究其可能的机制。方法随机将SD大鼠分为假手术组、脑瘫( CP)模型未给药组、CP模型左归丸组、CP模型巴氯芬组以及CP模型左归丸+巴氯芬联用组。分别于药物处理前,处理7、14、21、28天及治疗结束后7天时分别测定各组大鼠神经行为学指标,治疗结束后7天后测定血清中肿瘤坏死因子( TNF-α),一氧化氮合酶( NOS)和脑组织中丙二醛( MDA)、超氧化物歧化酶( SOD)的变化。结果巴氯芬治疗组和联用组在服药14天后与同期CP模型未给药组比较肌张力显著降低(t值分别为4.603、5.590,均P<0.05),通过平衡木时间缩短(t值分别为6.337、5.575,均P<0.05);而左归丸组于治疗28天后,大鼠通过平衡木时间才显著缩短(t=6.124,P<0.05)。左归丸治疗组和联用组大鼠服药28天后学习记忆能力较CP模型未给药组明显改善( t值分别为-4.314、-5.314,均P<0.05);联用组效果优于左归丸组(t=-2346,P<0.05);而巴氯芬处理组大鼠则无明显变化(t=-2.025,P>0.05)。与CP模型未给药组相比,左归丸组以及联用组大鼠的TNF-α、NOS和MDA显著降低,SOD显著升高,同时神经生长因子( NGF)和脑源性神经营养因子(BDNF) mRNA和蛋白水平升高(左归丸组:t 值分别为2.456、4.898、5.803、-6.144,-4.224、-5.124、-3.272、-4.166;联用组:t值分别为2.833、5.009、9.078、-6.056,-7.439、-7.142、-5.812、-4.301;均P<0.05),联用组的作用比左归丸组更强(t值分别为2.613、3.241、2.624、-3.194、-3.275、-4.002、-3.986、-2.881,均P<0.05),而巴氯芬组上述指标无明显变化(P>0.05)。结论左归丸和巴氯芬联用与单独用药相比能明显改善大鼠运动,且有可能通过保护神经细胞,促进神经元的修复来改善其认知功能。

Objective To investigate the effect of the combined application of Zuoguiwan and Baclofen on motor coordination ability and cognitive function of newborn rats with cerebral palsy ( CP) and to study its possible mechanism.Methods SD rats were randomly divided into sham-operated group, CP model without treatment group, CP model with Zuoguiwan group, CP model with Baclofen group, and CP model with Zuoguiwan combined with Baclofen group.Neurobehavioral indexes were detected in each group before treatment, 7, 14, 21 and 28 day treatment, and 7 day after treatment, respectively.TNF-αand NOS in serum, and MDA and SOD in brain tissue of rats were measured in each group 7 day after treatment.Results Compared with CP model without treatment group, muscle tension reduced significantly and the time of going through the balance reduced in Baclofen group and Zuoguiwan combined with Baclofen group after having taken the drugs for 14 days (t value was 4.603, 5.590, 6.337 and 5.575, respectively, all P<0.05).While Zuoguiwan group shortened the time to go through the balance after 28 day treatment (t=6.124,P<0.05).Compared with CP model without treatment group, the contents of TNF-α, NOS and MDA declined significantly but the levels of SOD, NGF and mRNA rose remarkably ( tZuoguiwan group value was 2.456, 4.898, 5.803, -6.144, -4.224, -5.124, -3.272 and -4.166, respectively, tcombined group value was 2.833, 5.009, 9.078,-6.056, -7.439, -7.142, -5.812 and -4.301, respectively, all P<0.05).The efficacy of the combined group was greater than Zuoguiwan group (t value was 2.613, 3.241, 2.624, -3.194, -3.275, -4.002, -3.986 and -2.881, respectively, all P<0.05).However, there was no obvious change in Baclofen group (P>0.05).Conclusion Compared with single drug therapy, combined application of Zuoguiwan and Baclofen can improve motor more obviously, and it may improve the cognition function of rats by protecting neurocyte and repairing neuron.
