目的:观察益气通脉胶囊对脑缺血损伤后大鼠脑微血管内皮细胞的r-谷氨酰转移酶(γ-GT)活性和血清血管内皮细胞生长因子( VEGF)表达的影响。方法大鼠采用反复夹闭双侧颈总动脉结合硝普钠降压法复制SD大鼠拟血管性痴呆模型。造模后选存活大鼠30只随机分为模型组、脑心通组和益气通脉胶囊组高中低剂量组,并设空白对照组。在给药第2天和第7天时尾静脉取血,分离血清,测定VEGF含量。连续给药20 d后分离大鼠大脑皮质的微血管内皮细胞,提取总蛋白,测定γ-GT的活性。结果微血管内皮细胞γ-GT的活性:给药20 d后,益气通脉胶囊中低剂量组大鼠的γ-GT的活性明显高于模型组。血清VEGF的含量:益气通脉胶囊各组的VEGF的含量在给药2d时与模型组相比差异无统计学意义,在给药7d时均明显高于模型组。结论益气通脉胶囊对脑缺血所致大鼠血脑屏障的损伤具有修复功能。

목적:관찰익기통맥효낭대뇌결혈손상후대서뇌미혈관내피세포적r-곡안선전이매(γ-GT)활성화혈청혈관내피세포생장인자( VEGF)표체적영향。방법대서채용반복협폐쌍측경총동맥결합초보납강압법복제SD대서의혈관성치태모형。조모후선존활대서30지수궤분위모형조、뇌심통조화익기통맥효낭조고중저제량조,병설공백대조조。재급약제2천화제7천시미정맥취혈,분리혈청,측정VEGF함량。련속급약20 d후분리대서대뇌피질적미혈관내피세포,제취총단백,측정γ-GT적활성。결과미혈관내피세포γ-GT적활성:급약20 d후,익기통맥효낭중저제량조대서적γ-GT적활성명현고우모형조。혈청VEGF적함량:익기통맥효낭각조적VEGF적함량재급약2d시여모형조상비차이무통계학의의,재급약7d시균명현고우모형조。결론익기통맥효낭대뇌결혈소치대서혈뇌병장적손상구유수복공능。
Objective To observe the effects of Yiqitongmai Capsule on the activity of Gamma Glu-tamyl Transpeptidase(γ - GT )in cerebral microvessels and the expression of Vascular Endothelial Cell Growth Factor( VEGF)in serum of rats with cerebral ischemic injury. Methods Healthy SD rats were ran-domized into control group and vascular dementia models,which were duplicated by repeated clamping of bi-lateral common carotid artery combined with sodium nitropruside depressing. Then models were divided into 5 groups:model group,Naoxintong( NXT)group and experimental groups( Yiqitongmai Capsule)including high, medium and low dose. On the second and sevenrth day of experiment,the concentrations of serum VEGF in rat were measured. After 20 days of continuous administration,cerebral cortices of rat were dissected and en-dothelia-enriched fractions were depleted,then extract the total protein and detect the activity of γ-GT. Results (1)The activities of γ-GT in the Yiqitongmai Capsule groups( low dose and medium dose)were higher than that in the model after 20 days of administration.(2)The concentrations of serum VEGF of all the experimental groups was higher than those of the model after 7 day of administration,while they seem to be similar at the beginning. Conclusions Yiqitongmai Capsule is effective on the repairing cerebral microves-sels in rats with cerebral ischemic injury.
