目的:建立大鼠长春瑞滨外渗性组织损伤的理想实验模型,在此动物模型上研究复脉膏外敷对外渗性损伤组织中VEGF 的影响,以探讨复脉膏外敷治疗化疗性静脉炎的疗效及机制。方法:42只大鼠随机分为2组:复脉膏组、50%硫酸镁组,21只/组,建立实验模型。2组大鼠左侧损伤处分别予复脉膏外敷、50%硫酸镁湿敷;右侧予生理盐水外敷作为对照组。在损伤后第1、4、7、11、14、18天,分别随机取3只大鼠,切除损伤部位的皮肤及皮下组织,固定标本,脱水、石蜡包埋,连续切片,采用免疫组化标记及图像分析方法,在电子显微镜下观察 VEGF 的表达情况。结果:VEGF 阳性表达主要见于表皮细胞、血管内皮细胞、毛囊上皮细胞胞浆、渗出的巨噬细胞、增生的成纤维细胞胞浆。复脉膏外敷较硫酸镁湿敷及生理盐水外涂能明显刺激长春瑞滨外渗性损伤组织中 VEGF 的阳性表达(P <0.01)。结论:复脉膏外敷于损伤皮肤后能上调损伤组织中 VEGF 的表达,通过 VEGF 的生物学功能,进而促进损伤组织的愈合。因此,复脉膏可以用于防治化疗性静脉炎。

목적:건립대서장춘서빈외삼성조직손상적이상실험모형,재차동물모형상연구복맥고외부대외삼성손상조직중VEGF 적영향,이탐토복맥고외부치료화료성정맥염적료효급궤제。방법:42지대서수궤분위2조:복맥고조、50%류산미조,21지/조,건립실험모형。2조대서좌측손상처분별여복맥고외부、50%류산미습부;우측여생리염수외부작위대조조。재손상후제1、4、7、11、14、18천,분별수궤취3지대서,절제손상부위적피부급피하조직,고정표본,탈수、석사포매,련속절편,채용면역조화표기급도상분석방법,재전자현미경하관찰 VEGF 적표체정황。결과:VEGF 양성표체주요견우표피세포、혈관내피세포、모낭상피세포포장、삼출적거서세포、증생적성섬유세포포장。복맥고외부교류산미습부급생리염수외도능명현자격장춘서빈외삼성손상조직중 VEGF 적양성표체(P <0.01)。결론:복맥고외부우손상피부후능상조손상조직중 VEGF 적표체,통과 VEGF 적생물학공능,진이촉진손상조직적유합。인차,복맥고가이용우방치화료성정맥염。
Objective:To establish an ideal animal experiment model of vinorelbine-induced extravasation injury to study effects of Fumai Paste on VEGF of tissue,and explore the curative effect and mechanism of treating chemotherapeutic phlebitis with external application of Fumai Paste.Methods:Forty-two rats were randomly divided into two groups with 21 rats in each group.Each rat was established to experimental model.Rats in Fumai Paste group were treated with external application of Fumai Paste on left side;and rats in magnesium sulfate groiup were treated with hydropathic compress of 50% magnesium sulfate on left side.The right side of rats in both groups were treated with hydropathic compress of normal saline.Three rats were selected randomly from each group on day 1,day 4,day 7,day 11,day 14 and day 18,and the skin and subcutaneous tissue of extravasation injury of each rat was cut off.All specimens were fixed in formalin,dehydrated,embedded in paraffin and serially cut.All specimens were observed expres-sions of VEGF under the electron microscope by immunohistochemical assay and image analysis method.Results:The positive ex-pression of VEGF was major in epidermic cells,vascular endothelial cell,endochylema of hair follicle cells,macrophage,endochyle-ma of fibroblast.Fumai Paste could stimulate the positive expression of VEGF tissue of animal experiment model with vinorelbine-induced extravasation injury more than 50% magnesium sulfate and normal saline (P <0.01).Conclusion:Fumai Paste can up-regulate the expression of VEGF tissue of animal experiment model with vinorelbine-induced extravasation injury.With the biologi-cal function of VEGF,Fumai Paste can accelerate the healing of the damaged tissues.Hence,chemotherapeutic phlebitis can be prevented and treated with external application of Fumai Paste.
