目的:研究老年人群中甾醇调节因子结合蛋白裂解激活蛋白(SCAP)单核苷酸多态性,分析基因型及血脂水平与高脂血症之间的关联性。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的分析方法,检测66例老年高脂血症患者(高脂血症组)和100例老年体检健康者(对照组)的SCAP基因Ile796Val的等位基因频率。结果高脂血症组、对照组等位基因A的频率分别为0.48、0.32,等位基因G的频率分别为0.52、0.68。高脂血症组与对照组之间的基因型和等位基因的频率差异有统计学意义(P<0.05)。结论 SCAP基因单核苷酸多态性导致脂质代谢紊乱,等位基因A可能是高脂血症的一个危险因子。

목적:연구노년인군중치순조절인자결합단백렬해격활단백(SCAP)단핵감산다태성,분석기인형급혈지수평여고지혈증지간적관련성。방법채용취합매련반응-한제성편단장도다태성(PCR-RFLP)적분석방법,검측66례노년고지혈증환자(고지혈증조)화100례노년체검건강자(대조조)적SCAP기인Ile796Val적등위기인빈솔。결과고지혈증조、대조조등위기인A적빈솔분별위0.48、0.32,등위기인G적빈솔분별위0.52、0.68。고지혈증조여대조조지간적기인형화등위기인적빈솔차이유통계학의의(P<0.05)。결론 SCAP기인단핵감산다태성도치지질대사문란,등위기인A가능시고지혈증적일개위험인자。
Objective To research single nucleotide polymorphism in SREBP cleavage-activating protein (SCAP) gene of senile hyperlipidaemia patients, and to analyze correlation in gene type, blood lipid level, and hyperlipidaemia. Methods Polymerase chain reaction-restricted fragment length polymorphisms (PCR-RFLP) was applied to detect allele frequency of Ile796Val in SCAP gene of 66 senile hyperlipidaemia patients (hyperlipidaemia group) and 100 healthy old people (control group). Results Frequency of allele A in hyperlipidaemia group and control group were respectively 0.48 and 0.32, and that of allele C were 0.52 and 0.68. There were statistical significant differences of gene type and allele frequency between hyperlipidaemia group and control group (P<0.05). Conclusion Single nucleotide polymorphism of SCAP gene will cause lipid metabolism disorders, and allele A may be one of the high risk factors of hyperlipidaemia.
