Monocrotaline Pulmonary%Arterial Hypertension%Rats
目的:观察脂氧素受体激动剂BML-111对野百合碱(MCT)诱导肺动脉高压大鼠的作用及其机制。方法选择SD大鼠24只,随机分为三组:C组、pH组、B组,pH组和B组大鼠行皮下注射野百合碱50μg/g,C组注射等量生理盐水,B组腹腔注射BML-1111mg/(kg·d),连续4周。观察平均肺动脉压力(MPAP)、右心室收缩压(RVSP)、右心室(RV)/[左室(LV)+室间隔(S)]、肺组织白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平的变化。结果 pH组、B组与C组比较,MPAP、RVSP、RV/(LV+S)、肺组织IL-6及TNF-α水平高于C组(P<0.05);B组与pH组比较,MPAP、RVSP、RV/(LV+S)、肺组织IL-6及TNF-α低于pH组(P<0.05)。结论脂氧素受体激动剂BML-111可减轻野百合碱诱导肺动脉高压大鼠肺动脉压力,其机制可能与减轻肺组织IL-6及TNF-α水平,抑制肺组织炎症有关。

목적:관찰지양소수체격동제BML-111대야백합감(MCT)유도폐동맥고압대서적작용급기궤제。방법선택SD대서24지,수궤분위삼조:C조、pH조、B조,pH조화B조대서행피하주사야백합감50μg/g,C조주사등량생리염수,B조복강주사BML-1111mg/(kg·d),련속4주。관찰평균폐동맥압력(MPAP)、우심실수축압(RVSP)、우심실(RV)/[좌실(LV)+실간격(S)]、폐조직백개소-6(IL-6)화종류배사인자-α(TNF-α)수평적변화。결과 pH조、B조여C조비교,MPAP、RVSP、RV/(LV+S)、폐조직IL-6급TNF-α수평고우C조(P<0.05);B조여pH조비교,MPAP、RVSP、RV/(LV+S)、폐조직IL-6급TNF-α저우pH조(P<0.05)。결론지양소수체격동제BML-111가감경야백합감유도폐동맥고압대서폐동맥압력,기궤제가능여감경폐조직IL-6급TNF-α수평,억제폐조직염증유관。
Objective To investigate the effect of BML-111 on rats pulmonary arterial hypertension induced by monocrotaline and mechanisms . Methods 24SD rats were randomly allocated into three groups: C group,pH group and B group,Both pH group and B group processed subcutaneous injection of 50μg/g monocrotaline while C group received same volume of saline,B group performed intraperitoneal injection of BML-111 1mg/kg/d from the day immediately as monocrotaline was injected. The observation: MPAP,RVSP,RV/(LV+S),the pulmonary tissue IL-6 and TNF-α. Results The pH group and B group developed higher level of MPAP,RVSP,RV/(LV+S),the pulmonary IL-6 and TNF-αas compared with C group(P<0.05),B group got lower increase of MPAP,RVSP,RV/(LV+S),the pulmonary IL-6 and TNF-αas compared with pH group(P<0.05). Conclusions BML-111 can ameliorate pulmonary arterial hypertension,inhibition of the pulmonary tissue inflammation may be a potential cause.
