目的:研究骨髓间充质干细胞在缺血条件下分泌的细胞因子浓度变化。方法用全骨髓差异贴壁法分离培养骨髓间充质干细胞(BMSCs),取第3代细胞进行实验。实验分两组:A组提取正常培养细胞的上清液;B组将细胞置于无血清的培养基中24h后,提取上清液。采用蛋白分析技术检测细胞培养上清液中27种细胞因子变化情况。结果无血清细胞培养组中ICAM-1、IFN-γ和IL-10的浓度与有血清的细胞培养组比较差异有统计学意义(P<0.05)。结论骨髓间充质干细胞在缺血条件下分泌的炎症相关细胞因子发生变化。

목적:연구골수간충질간세포재결혈조건하분비적세포인자농도변화。방법용전골수차이첩벽법분리배양골수간충질간세포(BMSCs),취제3대세포진행실험。실험분량조:A조제취정상배양세포적상청액;B조장세포치우무혈청적배양기중24h후,제취상청액。채용단백분석기술검측세포배양상청액중27충세포인자변화정황。결과무혈청세포배양조중ICAM-1、IFN-γ화IL-10적농도여유혈청적세포배양조비교차이유통계학의의(P<0.05)。결론골수간충질간세포재결혈조건하분비적염증상관세포인자발생변화。
Objective To study the secreted cytokine concentration changes of bone marrow mesenchymal stem cells(BMSCs)in ischemic conditions. Methods Bone marrow mesenchymal stem cells were cultured by adherent culture of whole bone marrow.The third generation of cells were used in this study. The experiments were divided into two groups:A. The supernatant wwas collected from normal cell culture. B. The supernatant was from cells in serum-free medium 24h. 27 kinds of cytokines in the supernatant were analyzed through the protein assay. Results Compared with the normal culture condition group,the concentration of ICAM-1,IFN-γand IL-10 in the serum-free culture group were significant decreased.(P<0.05). Conclusion There were changes in the inflammation related cytokines secreted by bone marrow mesenchymal stem cells in the ischemic condition.
