目的:探讨高糖状态对体外培养血管内皮细胞迁移、增殖能力的影响,以及其潜在可能的分子机理。方法:通过不同质量分数的葡萄糖干预血管内皮细胞后,用CCK-8评估内皮细胞增殖情况;用transwell实验检测内皮细胞的迁移能力;用Real -time PCR检测腺苷酸环化酶(adenylate cyclase,AC)mRNA的表达;用Western blotting 检测PKA、p-PKA蛋白的表达情况。结果:高质量分数葡萄糖干预血管内皮细胞后,血管内皮细胞迁移、增殖能力明显下降,同时AC mRNA表达明显下降,PKA蛋白表达无明显变化,而p-PKA蛋白表达明显下降。结论:高质量分数葡萄糖使血管内皮细胞迁移、增殖能力下降,cAMP-PKA信号通路的活化可能参与了血管内皮细胞增殖、迁移及修复功能。

목적:탐토고당상태대체외배양혈관내피세포천이、증식능력적영향,이급기잠재가능적분자궤리。방법:통과불동질량분수적포도당간예혈관내피세포후,용CCK-8평고내피세포증식정황;용transwell실험검측내피세포적천이능력;용Real -time PCR검측선감산배화매(adenylate cyclase,AC)mRNA적표체;용Western blotting 검측PKA、p-PKA단백적표체정황。결과:고질량분수포도당간예혈관내피세포후,혈관내피세포천이、증식능력명현하강,동시AC mRNA표체명현하강,PKA단백표체무명현변화,이p-PKA단백표체명현하강。결론:고질량분수포도당사혈관내피세포천이、증식능력하강,cAMP-PKA신호통로적활화가능삼여료혈관내피세포증식、천이급수복공능。
Objective:To determine the influence and its molecular mechanism of endothelial cell migration and proliferation in high glucos concentrations.Methods:The endothelial cells were treated in different glucose concentrations;and evaluated cell proliferation with CCK-8 reagent;and evaluated endothelial cell migration with transwell;and detected adenylate cyclase ( AC adenylate cyclase ) mRNA expression with fluorescence quantitative PCR;and detected PKA、p-PKA expression with Western blotting.Results:After endothelial cells were treated in high glucose concentrations , endothelial cell migration and proliferation significantly decreased ,and AC mRNA expression decreased obviously ,PKA of protein expression was not obviously changed ,and p-PKA expression decreased obviously.Conclusion:After endothelial cells were treated in high glucose concentrations , endothelial cell migration and proliferation are significantly decreased ,and cAMP-PKA activation may be involved in endothelial cell proliferation ,migration and repair functions.
