在单因素的基础上,选取酶浓度、酶解温度、酶解时间、pH四个因素为自变量,以脱胶后油脂中的磷含量为响应值,根据中心组合设计原理,利用Design–Expert 7.0软件设计响应面试验及对数据进行处理,优化酶解工艺,确定最佳工艺参数为:酶浓度0.045%、反应时间3.5 h、反应温度44.0℃、pH为4.8。在此条件进行磷脂酶Lecitase Ultra脱除大豆油中非水化磷脂实验,3次平行实验所得到的磷含量的平均值为6.2 mg/kg。

The influence of Lecitase Ultra on removal of nonhydratable phospholipids in soybean oil was studied. On the basis of single–factor tests,enzyme concentration,reaction temperature,reaction time and the pH were selected as independent variables,and the phosphorus content in degummed oil was selected as response value. The test was designed to use the response surface methodology according to central composite design principle. The software Expert Design 7.0 Trial was used for data processing. The results showed that the optimum conditions were enzyme concentration 0.045%, reaction time 3.5 h,reaction temperature 44.0℃ and pH 4.8. According to this condition,the experiment of removing the nonhydratable phospholipids in soybean oil by Lecitase Ultra was made, and the average phosphorus content got from three parallel experiments was 6.2 mg/kg.
