目的:本文拟建立成熟慢性病毒性肝炎的动物模型,探讨赶黄草与黄芩不同配比对病毒性肝炎的影响,为临床用药提供实验依据。方法建立C3H/HeJ小鼠病毒性肝炎模型,采用试剂盒检测模型组、赶黄草组、黄芩组、赶黄草配黄芩1∶1组、1∶2组、1∶3组各给药组小鼠血清ALT、AST、STB、TP指标的含量变化以及采用光学显微镜观察模型组和各给药组小鼠肝脏的形态学改变;采用免疫组织化学图像分析法测定各组肝组织COX-2的表达。结果与空白组比较,模型组的肝组织呈炎性病理改变,肝脏指数显著降低,血清中ALT、AST、STB含量显著升高,TP含量显著下降。与模型组比较,赶黄草组、赶黄草配黄芩1∶1组、1:2组、1∶3组小鼠肝组织炎症、纤维化等明显减轻,血清中的AST、ALT、STB含量显著降低;赶黄草组、赶黄草配黄芩1∶1组、1∶2组血清TP含量显著升高。COX-2的表达结果显示,空白组肝细胞仅见少量阳性表达,与空白组比较,模型组肝组织中COX-2的表达显著增强;与模型组比较,各治疗组肝组织中COX-2表达均明显低于模型组,其中赶黄草配黄芩1:2组的表达最弱。结论赶黄草能显著改善小鼠肝炎的病变,黄芩有改善其炎性病变的趋势,二者配伍后,尤其当黄芩2倍于赶黄草时,能显著缓解其肝炎病变,其机制可能与降低COX-2的表达有关。提示临床治疗病毒性肝炎用药,黄芩2倍用量于赶黄草效果最佳。

목적:본문의건립성숙만성병독성간염적동물모형,탐토간황초여황금불동배비대병독성간염적영향,위림상용약제공실험의거。방법건립C3H/HeJ소서병독성간염모형,채용시제합검측모형조、간황초조、황금조、간황초배황금1∶1조、1∶2조、1∶3조각급약조소서혈청ALT、AST、STB、TP지표적함량변화이급채용광학현미경관찰모형조화각급약조소서간장적형태학개변;채용면역조직화학도상분석법측정각조간조직COX-2적표체。결과여공백조비교,모형조적간조직정염성병리개변,간장지수현저강저,혈청중ALT、AST、STB함량현저승고,TP함량현저하강。여모형조비교,간황초조、간황초배황금1∶1조、1:2조、1∶3조소서간조직염증、섬유화등명현감경,혈청중적AST、ALT、STB함량현저강저;간황초조、간황초배황금1∶1조、1∶2조혈청TP함량현저승고。COX-2적표체결과현시,공백조간세포부견소량양성표체,여공백조비교,모형조간조직중COX-2적표체현저증강;여모형조비교,각치료조간조직중COX-2표체균명현저우모형조,기중간황초배황금1:2조적표체최약。결론간황초능현저개선소서간염적병변,황금유개선기염성병변적추세,이자배오후,우기당황금2배우간황초시,능현저완해기간염병변,기궤제가능여강저COX-2적표체유관。제시림상치료병독성간염용약,황금2배용량우간황초효과최가。
Objective To construct the mice model of chronic virus hepatitis and to study the influence of Penthorum,Penthorum and Scutellariae with different proportion on the model of chronic virus hepatitis. Methods All the mice were divided into Penthorum group,scutellariae group,Penthorum with Scutellariae 1∶1group,1∶2 group and 1∶3 group seven different groups,then established the model of C3H/HeJ mice institute of viral hepatitis;Taking use of kits to detect the content of changes in groups’ mice serum ALT,AST,STB,of TP index and the morphological changes of mice liver;Using the immunohistochemical image method to detect the expression of cox-2. Results Compared with the blank group,the liver tissue of model group had inflammatory pathological change,the liver index decreased significantly,and there were significant elevated in serum ALT,AST,STB,while TP content decreased significantly;Compared with model group,ALT,AST,STB content in mice serum of Penthorum group,Penthorum with Scutellariae 1:1group,1∶2 group and 1∶3 group decreased significantly;in the study of TP, the content of Penthorum group,Penthorum with Scutellariae 1∶1 group,1∶2 group increased significantly. The research of expression of COX-2 showed that,the expression of blank group had only a small amount of positive result;Compared with the blank group,the expression of COX-2 in liver tissue of model group was significantly enhanced;Compared with model group,the expression of COX-2 in treatment group were significantly lower,and the expression of Penthorum with Scutellariae 1∶2 group was the weakest. Conclusion Penthorum can significantly improve the symptoms of hepatitis in mice,Scutellariae has a tendency to improve its inflammatory symptoms,when both two compatible used,especially scutellaria 2 times than Penthorum,could significantly relieve the symptoms. During the clinical medication,dosage of Scutellariae twice than Penthorum could be better.
