目的 探讨大鼠棉花烟雾吸入性肺损伤中的氧化应激反应机制.方法 18只雄性SD大鼠随机分成对照组、6h组、24 h组,每组6只.复制大鼠棉花烟雾吸入性肺损伤模型,6h组、24 h组大鼠分别在烟雾吸入后6h或24 h行安乐死,ELISA法检测大鼠肺组织丙二醛(MDA)、谷胱甘肽(GSH)、一氧化氮(NO)、诱导型一氧化氮合酶(iNOS)、γ-谷氨酰半胱氨酸合成酶(γ-GCS)浓度,荧光定量PCR法行肺组织iNOS mRNA、γ-GCS mRNA定量.结果 6h组、24 h组大鼠肺组织匀浆中的MDA、NO、iNOS、γ-GCS浓度、iNOS mRNA和γ-GCS mRNA的相对表达量均高于对照组,且NO浓度、iNOSmRNA和γ-GCS mRNA的相对表达量24 h组高于6h组(P值均<0.05);而6h组和24 h组GSH浓度均低于对照组,且24 h组低于6h组(P值均<0.05).结论 在大鼠棉花烟雾吸入性肺损伤中,氧化应激反应加剧,同时抗氧化体系的激活不足以对抗氧化应激损伤,氧化/抗氧化体系失衡,导致肺损伤逐渐加重.

목적 탐토대서면화연무흡입성폐손상중적양화응격반응궤제.방법 18지웅성SD대서수궤분성대조조、6h조、24 h조,매조6지.복제대서면화연무흡입성폐손상모형,6h조、24 h조대서분별재연무흡입후6h혹24 h행안악사,ELISA법검측대서폐조직병이철(MDA)、곡광감태(GSH)、일양화담(NO)、유도형일양화담합매(iNOS)、γ-곡안선반광안산합성매(γ-GCS)농도,형광정량PCR법행폐조직iNOS mRNA、γ-GCS mRNA정량.결과 6h조、24 h조대서폐조직균장중적MDA、NO、iNOS、γ-GCS농도、iNOS mRNA화γ-GCS mRNA적상대표체량균고우대조조,차NO농도、iNOSmRNA화γ-GCS mRNA적상대표체량24 h조고우6h조(P치균<0.05);이6h조화24 h조GSH농도균저우대조조,차24 h조저우6h조(P치균<0.05).결론 재대서면화연무흡입성폐손상중,양화응격반응가극,동시항양화체계적격활불족이대항양화응격손상,양화/항양화체계실형,도치폐손상축점가중.
Objective To investigate the mechanisms of oxidative stress in cotton smoke inhalationinduced acute lung injury in rats.Methods Eighteen male SD rats were randomly divided into control group,6 h group,and 24 h group.The rat model of cotton smoke inhalation injury was established.After smoke inhalation rats were mercy killed.The concentrations of malondialdehyde (MDA),glutathione (GSH),nitric oxide (NO),inducible nitric oxide synthase (iNOS),and γ-glutamyl systeine synthetase (γ-GCS) in homogenized lung tissue were measured by enzyme-linked immunosorbent assay and the relative expressions of iNOS mRNA and γ-GCS mRNA were detected by fluorescence quantitative PCR.Results Compared with the control group,concentrations of MDA,NO,iNOS,γ-GCS and relative expressions of iNOS mRNA and γ-GCS mRNA in homogenized lung tissue in 6 h group and 24 h group were significantly elevated (all P <0.05),moreover,concentration of NO and relative expressions of iNOS mRNA and γ-GCS mRNA in 24 h group were higher than those in 6 h group (all P <0.05).The concentration of GSH in homogenized lung tissue in 6 h group was lower than that in control group,but higher than that in 24 h group.Conclusions Oxidative stress significantly enhances in rats with cotton smoke inhalation-induced acute lung injury,meanwhile,the activation of antioxidative system is not enough.As a result,the oxidative-antioxidative balance is disturbed and lung injury becomes worse.
