目的:探讨用RNAi介导瘦素受体基因(ObR)沉默人乳头状甲状腺癌K1细胞(K1细胞)增殖与侵袭能力的影响。方法使用 siRNA 干扰技术瞬时转染构建 siRNA‐ObR。瞬时转染后,Real‐time PCR和Western blotting在mRNA水平和蛋白水平检测siRNA‐ObR是否有效抑制K1细胞内靶基因的表达;MTT 实验研究ObR沉默后K1细胞的增殖能力;Transwell实验检测ObR沉默后K1细胞的侵袭能力。结果 Real‐time PCR实验结果显示,ObR基因沉默后K1细胞ObR mRNA表达显著降低;Western blotting实验结果显示, ObR基因沉默后K1细胞ObR蛋白表达显著降低;M T T实验结果显示ObR对K1细胞的增殖没有意义;T ran‐swell实验结果显示ObR沉默后K1细胞侵袭能力显著下降。结论 ObR基因沉默能有效抑制K1细胞ObR基因的表达,降低K1细胞的侵袭能力。

목적:탐토용RNAi개도수소수체기인(ObR)침묵인유두상갑상선암K1세포(K1세포)증식여침습능력적영향。방법사용 siRNA 간우기술순시전염구건 siRNA‐ObR。순시전염후,Real‐time PCR화Western blotting재mRNA수평화단백수평검측siRNA‐ObR시부유효억제K1세포내파기인적표체;MTT 실험연구ObR침묵후K1세포적증식능력;Transwell실험검측ObR침묵후K1세포적침습능력。결과 Real‐time PCR실험결과현시,ObR기인침묵후K1세포ObR mRNA표체현저강저;Western blotting실험결과현시, ObR기인침묵후K1세포ObR단백표체현저강저;M T T실험결과현시ObR대K1세포적증식몰유의의;T ran‐swell실험결과현시ObR침묵후K1세포침습능력현저하강。결론 ObR기인침묵능유효억제K1세포ObR기인적표체,강저K1세포적침습능력。
Objective To study the effect of RNAi -mediated OBR gene silencing on human papillary thyroid carcinoma cell K1 . Methods siRNA interference technology transient transfection was used to construction of siRNA-ObR .After transfection ,we detected whether siRNA‐ObR inhibited the expression of target genes in mR‐NA level and protein level .MTT and Transwell respectively detected proliferation and invasion of K 1 cells after OBR gene knockdown .Results Real‐time PCR results showed that the level of mRNA expression decreased in K 1 cells after ObR gene silencing .Western Blotting results exhibited that the level of protein expression was inhibited by ObR gene silencing .MTT results showed the silencing of ObR did not influence the proliferation of K 1 cells . Transwell results demonstrated the silencing of ObR decreased K 1 cells invasion .Conclusion OBR siRNA could effectively inhibit the expression of target gene and reduce invasive ability of human papillary thyroid carcinoma cell K1 .
