Severe acute pancreatitis%Tanreqing%Pulmonary injury%Rats
目的:观察痰热清注射液对重症急性胰腺炎大鼠肺损伤的保护作用。方法选择96只健康SD大鼠随机分为对照组、假手术组、模型组及痰热清组,采用5%牛磺胆酸钠逆行胰胆管注射法建立SAP大鼠模型,以建模后6、12 h及24 h,分为3个亚组(每组8只)。观察肺损伤指数,分别采用酶联免疫法、半定量逆转录PCR法及蛋白印迹技术检测不同时间点肺组织中RNA及高迁移率族蛋白水平。结果模型组较对照组和假手术在肺损伤指数、细胞间粘附分子-1(ICAM-1)、白介素-18(IL-18)、骨髓系细胞触发受体(TREM-1) mRNA及高迁移率族蛋白-1(HMGB1)方面差异均有统计学意义(P<0.01),而痰热清组治疗后较模型组ICAM-1、IL-18、TREM-1 mRNA和HMGB1等水平有降低(P<0.01),肺损伤指数明显降低(P<0.01)。结论痰热清注射液能降低重症急性胰腺炎大鼠肺组织中ICAM-1、IL-18、TREM-1 mRNA及高迁移率族蛋白B1水平的表达,有效缓解重症急性胰腺炎大鼠的肺损伤。

목적:관찰담열청주사액대중증급성이선염대서폐손상적보호작용。방법선택96지건강SD대서수궤분위대조조、가수술조、모형조급담열청조,채용5%우광담산납역행이담관주사법건립SAP대서모형,이건모후6、12 h급24 h,분위3개아조(매조8지)。관찰폐손상지수,분별채용매련면역법、반정량역전록PCR법급단백인적기술검측불동시간점폐조직중RNA급고천이솔족단백수평。결과모형조교대조조화가수술재폐손상지수、세포간점부분자-1(ICAM-1)、백개소-18(IL-18)、골수계세포촉발수체(TREM-1) mRNA급고천이솔족단백-1(HMGB1)방면차이균유통계학의의(P<0.01),이담열청조치료후교모형조ICAM-1、IL-18、TREM-1 mRNA화HMGB1등수평유강저(P<0.01),폐손상지수명현강저(P<0.01)。결론담열청주사액능강저중증급성이선염대서폐조직중ICAM-1、IL-18、TREM-1 mRNA급고천이솔족단백B1수평적표체,유효완해중증급성이선염대서적폐손상。
Objective:To observe the protective effect of Tanreqing Injection on pulmonary injury by severe acute pancreatitis in rats. Methods:96 healthy SD rats were randomly divided into control group,sham opera?tion group,model group and Tanreqing group,5% sodium taurocholate retrograde cholangiopancreatography injec?tion rat model was established by SAP to modeling after 6 h,12 h and 24 h,divided into three sub-groups(n=8). Indicators of lung injury,and lung tissue ICAM-1,IL-18,TREM-1mRNA were observed and high mobility group protein B1 level was detected by enzyme-linked immunosorbent assay,semi-quantitative RT-PCR and Western blot at different time points respectively. Results:The lung injury index,ICAM-1,IL-18 TREM-1mRNA and HMGB1 of the model group had significant differences from the control group and sham operation group(P< 0.01),but after Tanreqing treatment group compared with the model group,IL-18,TREM-1mRNA and HMGB1 levels were lower(P<0.01),Pulmonary injury parameters were significantly lower(P<0.01). Con?clusion:Tanreqing can decrease the expression of the ICAM-1,IL-18,TREM-1mRNA and high mobility group protein B1 in severe acute pancreatitis rats,and effectively alleviate lung injury in rats with the severe acute pan?creatitis.
