目的:探讨内吗啡肽在视网膜的分布,内吗啡肽和μ阿片受体在视网膜的共存关系。方法成年雄性 SD 大鼠12只,分为2组,一组为正常对照组(n =6),另一组建立高眼压模型(高眼压模型组,n =6)。运用免疫组织化学方法观察内吗啡肽1(EM-1)、内吗啡肽2(EM-2)和μ阿片受体在视网膜中的分布和共存,并利用高眼压模型观察高眼压对 EM-2在视网膜表达的影响。结果视网膜无 EM-1分布;EM-2主要分布于视网膜节细胞层;EM-2与 NeuN、μ阿片受体在视网膜节细胞层共存;高眼压模型大鼠视网膜节细胞层内单位面积表达 EM-2的细胞数明显少于正常大鼠(0.8±0.1个/mm3 vs.4.6±0.8个/mm3),差异有统计学意义(t =3.88,P<0.05)。结论EM-2分布在视网膜节细胞层,与μ阿片受体共存,在高眼压状态下,EM-2的表达减少。

Objective To investigate the distribution of endomorphins (EM)in the retina,and as-sess the coexistence of EMand μ-opioid receptor in the retina.Methods Twelve Sprague Dawley (SD)rats were divided into the control (n =6)and intraocular hypertension model groups (n =6).Immunohistochemis-try was used to detect the expression and coexistence of EM-1,EM-2 and μ-opioid receptor in the rat retina, and evaluate the influence of intraocular hypertension upon the expression of EM-2.Results EM-1 was not expressed in the retina.EM-2 was mainly distributed in the retinal ganglion cell layer.EM-2,NeuN and μ-opioid receptor coexisted in the retinal ganglion cell layer.The density of cells expressing EM-2 in the intraocu-lar hypertension model group was (0.8 ±0.1) /mm3 ,significantly lower compared with (4.6 ±0.8) /mm3 in the control group (t =3.88,P < 0.05).Conclusions EM-2 was expressed in the retinal ganglion cell layer and coexisted with μ-opioid receptor.The expression of EM-2 was decreased under intraocular hyperten-sion.
