目的：从皮肤组织块培养获取原代细胞。方法：将皮肤组织块修剪成2mm×2mm大小，放置到培养皿中，真皮层向下，观察细胞爬出的情况。结果：组织块培养第3天，有角质形成细胞从组织块爬出，第7天左右组织块周围出现黑素细胞，随着培养时间的延长，组织块周围出现大量成纤维细胞。结论：在组织块培养过程中，早期可以收集较纯的角质形成细胞，含血清的DMEM培养20d后，可以得到大量成纤维细胞。
목적：종피부조직괴배양획취원대세포。방법：장피부조직괴수전성2mm×2mm대소，방치도배양명중，진피층향하，관찰세포파출적정황。결과：조직괴배양제3천，유각질형성세포종조직괴파출，제7천좌우조직괴주위출현흑소세포，수착배양시간적연장，조직괴주위출현대량성섬유세포。결론：재조직괴배양과정중，조기가이수집교순적각질형성세포，함혈청적DMEM배양20d후，가이득도대량성섬유세포。
Objective To harvest primary cells from skin explants culture. Methods Skin explants in size of 2mm × 2mm were obtained by using sharp scissors,each skin explant placed onto the bottom of culture dish,and they were oriented with the epidermis facing up,then observe the cells migrating from the explants. Results From the third day,keratinocytes migrating from the explants,then melanocytes appear around the explants.As culture time getting longger,a large number of fibrolasts around the explant. Conclusion During the explant culture,we could harvest keratinocyte without contamination firstly,and DMEM contain serum culture for 20days,could harvest a large number of fibrolasts.
