目的:建立中药木瓜药材中总糖及还原糖的测定方法并对其含量进行测定,由此计算推测出药材中多糖含量。方法采用苯酚-浓硫酸法测定总糖含量,采用3,5-二硝基水杨酸法(DNS法)测定还原糖的含量。结果木瓜总糖含量为12.06%。DNS法测定还原糖的最佳条件为院测定波长560 nm,DNS最佳用量为5.0 mL,沸水浴加热5 min。测得药材中还原糖含量为11.58%。木瓜原药材中多糖含量为2.51%。结论苯酚浓硫酸法及DNS法可用于总糖及还原糖含量的测定,总糖和还原糖含量的高低在一定程度上可以反映多糖含量的多少。

Objective To establish a method for determination of total sugar and reducing sugar in Chaenomeles speciosa and to detect the content of sugar with the methods. Methods The content of total sugar was determined by phenol-sulfuric acid method. The concentration of reducing sugar was determined by 3, 5-dinitrosalicylic acid method. Results The content of total sugar in Chaenomeles speciosa was 12.06%. The optimum determination wavelength of DNS colorimetry was 560 nm. The proper volume of DNS sample was 5.0 mL. The best time for colour reaction in the condition of boiling water bath was kept for 5 min. The content of reducing sugar in Chaenomeles speciosa was 11.58%. The content of polysaccharides was 2.51%. Conclusion Phenol-sulfuric acid method and DNS method can be used for the determination of total sugar and reducing sugar. The content of total sugar and reducing sugar indicate the content of polysaccharides in some degree.
