以黄色短杆菌FY-18为出发菌株,利用摇瓶发酵生产L-亮氨酸,并利用正交试验和单因素试验分别对其发酵培养基和基本发酵条件进行研究,从而优选出最佳培养基组分和培养条件参数。结果表明, L-亮氨酸发酵培养基的最适配比为:葡萄糖160 g/L、豆粕水解液20 g/L、玉米浆20 g/L、毛发粉15 g/L、硫酸铵70 g/L;其发酵最佳培养条件为:初始pH 7.2,培养温度32℃,发酵接种量为10%。在上述最佳条件下摇瓶中发酵的产酸量为22.5 g/L。
以黃色短桿菌FY-18為齣髮菌株,利用搖瓶髮酵生產L-亮氨痠,併利用正交試驗和單因素試驗分彆對其髮酵培養基和基本髮酵條件進行研究,從而優選齣最佳培養基組分和培養條件參數。結果錶明, L-亮氨痠髮酵培養基的最適配比為:葡萄糖160 g/L、豆粕水解液20 g/L、玉米漿20 g/L、毛髮粉15 g/L、硫痠銨70 g/L;其髮酵最佳培養條件為:初始pH 7.2,培養溫度32℃,髮酵接種量為10%。在上述最佳條件下搖瓶中髮酵的產痠量為22.5 g/L。
이황색단간균FY-18위출발균주,이용요병발효생산L-량안산,병이용정교시험화단인소시험분별대기발효배양기화기본발효조건진행연구,종이우선출최가배양기조분화배양조건삼수。결과표명, L-량안산발효배양기적최괄배비위:포도당160 g/L、두박수해액20 g/L、옥미장20 g/L、모발분15 g/L、류산안70 g/L;기발효최가배양조건위:초시pH 7.2,배양온도32℃,발효접충량위10%。재상술최가조건하요병중발효적산산량위22.5 g/L。
Brevibacterium flavum FY-18 was used as original strain to produce L-leucine by shaking flask fermentation. The orthogonal experiments and single factor experiments were adopted to optimize the fermentation culture medium and conditions of L-leucine. The results indicated that the optimized compositions of medium were 160 g/L glucose, 20 g/L soybean meal hydrolysate, 20 g/L corn steep liquor, 15 g/L hair powder and 70 g/L ammonium sulfate, and the fermentation conditions were initial pH at 7.2, 32℃and inoculum 10%. Under the optimized fermentation conditions the production of L-leucine was up to 22.5 g/L .
